摘  要:试验旨在探究NR6A1基因在杜撒×大长撒猪中的多态性及其对育肥、胴体和肌肉品质的影响。试验共测定了321头杜撒×大长撒猪的育肥、胴体和肉质指标,采用PCR扩增产物直接测序法对杜撒×大长撒猪(n=321)、杜洛克(n=35)、长白(n=32)、大白(n=32)NR6A1基因10个外显子进行单核苷酸多态性检测并计算NR6A1基因多态位点基因型频率及基因频率,采用最小二乘模型分析各SNPs位点与育肥、胴体、肉质的关联。结果表明,杜撒×大长撒猪NR6A1基因第5外显子检测到一个SNP位点(NR6A1 g.265347265T>C),该位点存在TT、TC和CC 3种基因型,处于中度多态(PIC=0.3670),杂合度(H)为0.4842,处于Hardy-Weinberg平衡状态(P>0.05);在杜洛克、长白、大约克中,NR6A1 g.265347265T>C位点TT基因型频率为1,表现为单态;杜撒×大长撒猪NR6A1 g.265347265T>C位点TT基因型肋骨数(14.13±0.06对)显著多于CC基因型(13.94±0.08对,P<0.05),TC基因型胴体直长(98.54±0.57 cm)显著大于CC基因型(95.94±1.07 cm,P<0.05),TT基因型胴体瘦肉率(55.28±0.53%)显著高于CC基因型(53.16±0.80%,P<0.05)。综上,NR6A1 g.265347265T>C位点T等位基因能显著增加杜撒×大长撒猪肋骨数、胴体直长和瘦肉率,可作为撒坝猪优化选育的分子标记。This study was conducted to explore the polymorphism of NR6A1 gene and its association between fattening,carcass and meat quality performances in(Duroc×Saba)×[Yorkshire×(Landrace×Saba)].In this experiment,the fattening,carcass and meat quality performances of 321 DS×YLS were measured.The single nucleotide polymorphism of 10 exons of NR6A1 gene in DS×YLS(n=321),Duroc(n=35),Landrace(n=32)and Large White(n=32)were detected by direct sequencing of PCR amplification products.The genotypic frequency and allele frequency of NR6A1 gene polymorphic loci were calculated.The association between genotypes and fattening,carcass and meat quality performances were analyzed by the least square model.The results showed that a SNP locus(NR6A1 g.265347265T>C)was located on exon 5 of NR6A1 gene in DS×YLS and there were 3 genotypes(TT,TC and CC)at NR6A1 g.265347265T>C locus,which was moderate polymorphisms(PIC=0.3670).The heterozygosity was 0.4842 in this locus and the genotype distribution was in accordance with the Hardy-Weinberg equilibrium(P>0.05).The NR6A1 g.265347265T>C locus was homozygous in Duroc,Landrace and Large White,which was TT genotype,showing monomorphism.The rib numbers in the TT genotype(14.13±0.06 pairs)was significantly more than that in the CC genotype(13.94±0.08 pairs,P<0.05).The carcass straight length in the TC genotype(98.54±0.57 cm)was significantly longer than that in the CC genotype(95.94±1.07 cm,P<0.05).The lean meat rate in the TT genotype(55.28±0.53%)was significantly longer than that in the CC genotype(53.16±0.80%,P<0.05).These results indicate that the T allele of NR6A1 g.265347265T>C locus has significant effects on the rib numbers,carcass straight length and lean meat rate of DS×YLS(P<0.05),and could be used as a potential molecular marker for optimal breeding in Saba pigs.
