摘 要:试验旨在建立1种检测围产期母羊胰岛素(INS)敏感性的方法,探究酮病对围产期母羊能量平衡的影响。试验1选择3只围产期绒山羊母羊进行高胰岛素-正血糖钳夹处理,分别在灌注前后测定绒山羊血浆INS和血糖(GLU)含量。试验2选取与试验1处于同一牧场、相同围产期(产后7 d)的绒山羊母羊为研究对象,选取酮病羊[血浆β-羟丁酸(BHBA)含量≥1.6 mmol/L]和非酮病羊(血浆BHBA含量<0.8 mmol/L)各6只,颈静脉采血检测绒山羊血浆INS、GLU、BHBA、游离脂肪酸(NEFA)的含量。结果显示:与处理前期相比,试验处理期绒山羊母羊GLU含量无显著变化(P>0.05),血浆INS含量极显著升高(P<0.01)。整个试验过程中,试验处理期绒山羊母羊的血浆INS含量均高于处理前期。酮病组母羊血浆GLU、BHBA、NEFA含量显著高于非酮病组母羊(P<0.05),血浆INS含量显著低于非酮病组母羊(P<0.05)。研究表明,试验成功构建了利用高胰岛素-正血糖钳夹技术检测围产期母羊INS敏感性的方法。The study aimed to establish a method for detecting insulin(INS)sensitivity in periparturient ewes and to investigate the impact of ketosis on the energy balance of periparturient ewes.Experiment 1 selected three periparturient cashmere goats and subjected them to hyperinsulinemic-euglycemic clamp treatment.The plasma INS and glucose(GLU)levels of the cashmere goats were measured before and after infusion.Experiment 2 chose cashmere goats in the same periparturient period(seven days postpartum)as those in Experiment 1 from the same farm as the subjects.Six goats with ketosis[plasmaβ-hydroxybutyrate(BHBA)level≥1.6 mmol/L]and six goats without ketosis(plasma BHBA level<0.8 mmol/L)were selected.Blood samples were collected from the jugular vein to detect the plasma levels of INS,GLU,BHBA,and non-esterified fatty acids(NEFA).The results showed that compared with the pre-treatment period,the plasma GLU level of the cashmere goats during the treatment period did not change significantly(P>0.05),while the plasma INS level was extremely increased(P<0.01).Throughout the experiment,the plasma INS level during the treatment period were higher than those before treatment.The ketotic goats had significantly higher plasma GLU,BHBA,and NEFA levels than the non-ketotic goats(P<0.05),while their plasma INS level was significantly lower than those of the non-ketotic goats(P<0.05).The study indicates that the hyperinsulinemic-euglycemic clamp technique is successfully used to detect INS sensitivity in periparturient ewes .
