补元汤调控IL-6/STAT3信号通路对COPD大鼠气道黏液高分泌的影响

摘  要:目的:观察补元汤对慢性阻塞性肺疾病(COPD)大鼠气道黏液高分泌的影响,并从白细胞介素-6(IL-6)/信号转导及转录激活因子3(STAT3)信号通路初步探讨其机制。方法:将36只SD大鼠随机分为对照组,模型组,补元汤低、中、高剂量组,氨茶碱组,每组6只。采用卷烟烟雾暴露联合气道内注射脂多糖建立COPD大鼠模型,造模4周后开始补元汤干预,给药4周后取材。分别于造模4周后、给药4周后检测大鼠肺功能指标最大吸气流速(PIF)、潮气量(TV)、每分钟通气量(MV)、气道狭窄指数(Penh)。HE染色观察大鼠肺组织病理改变;PAS染色观察大鼠气道黏液高分泌程度,运用Image-Pro Plus 6.0图像软件计算支气管基底膜单位长度内杯状细胞数目;Real time-PCR和Western Blot检测肺组织IL-6、STAT3 mRNA及蛋白表达;免疫组化法检测肺组织黏蛋白(MUC)5AC、基质金属蛋白酶-9(MMP-9)、中性粒细胞弹性蛋白酶(NE)表达水平。结果:造模4周后,与对照组比较,模型组大鼠PIF、TV、MV均显著下降(P<0.01),Penh显著升高(P<0.01)。给药4周后,与模型组比较,补元汤各剂量组与氨茶碱组大鼠PIF、TV、MV均显著升高(P<0.01),Penh显著下降(P<0.01);肺组织病理损伤和气道黏液分泌程度明显减轻,单位长度内杯状细胞数目显著减少(P<0.01);补元汤中、高剂量组及氨茶碱组大鼠肺组织IL-6 mRNA表达显著下降(P<0.05),补元汤各剂量组及氨茶碱组STAT3 mRNA表达显著下降(P<0.01);补元汤各剂量组及氨茶碱组大鼠肺组织IL-6蛋白表达显著下降(P<0.05,P<0.01),补元汤中、高剂量组及氨茶碱组STAT3蛋白表达明显下降(P<0.05,P<0.01);MUC5AC、MMP-9、NE表达显著降低(P<0.01),其中补元汤高剂量组与氨茶碱组改善效果最接近。结论:补元汤可能通过调控IL-6/STAT3信号通路改善COPD大鼠气道黏液高分泌。Objective:To observe the effect of Buyuan Decoction on airway mucus hypersecretion in chronic obstructive pulmonary disease(COPD)rats,and to explore its mechanism through the interleukin-6(IL-6)/signal transducer and activator of transcription 3(STAT3)signaling pathway.Methods:A total of 36 SD rats were randomly divided into the control group,model group,low-,medium-,and high-dose Buyuan Decoction groups,and the aminophylline group,with 6 rats in each group.COPD models were established using a combination of cigarette smoke exposure and lipopolysaccharide(LPS)intra-airway injection.After 4 weeks of modeling,Buyuan Decoction intervention was started,and tissues were collected after 4 weeks of treatment.Pulmonary function indicators such as peak inspiratory flow(PIF),tidal volume(TV),minute ventilation volume(MV),and enhanced enhancedpause(Penh)were measured 4 weeks post-modeling and post-treatment.Histopathological changes in lung tissues were observed using hematoxylin-eosin(HE)staining;the degree of airway mucus hypersecretion was assessed using periodic acid-Schiff(PAS)staining,and the number of goblet cells per unit length of the bronchial basement membrane was calculated using the Image-Pro Plus 6.0 image software.Real-time PCR and Western blot were used to detect the expression of IL-6 and STAT3 mRNA and protein in lung tissues;immunohistochemistry was employed to examine the expression levels of mucin 5AC(MUC5AC),matrix metalloproteinase-9(MMP-9),and neutrophil elastase(NE)in lung tissues.Results:Four weeks after modeling,compared to the control group,the PIF,TV,and MV of the model group rats significantly decreased(P<0.01),while Penh significantly increased(P<0.01).After 4 weeks of treatment,compared to the model group,rats in all Buyuan Decoction dose groups and the aminophylline group showed significant increases in PIF,TV,and MV(P<0.01)and significant decreases in Penh(P<0.01).Pathological damage in lung tissues and the degree of airway mucus secretion were significantly alleviated,with a notable reduct
