天龙竭对博来霉素诱导的肺间质纤维化大鼠血管重构的影响

摘  要:目的:通过观察天龙竭对博来霉素诱导的肺间质纤维化大鼠血管重构的影响,探讨天龙竭抗肺间质纤维化的作用机制。方法:将60只Wistar大鼠随机分为空白组,模型组,天龙竭高、中、低剂量组,吡非尼酮组,每组10只。除空白组外,其余组均采用气管内滴注博来霉素法复制肺间质纤维化大鼠模型。造模后第7天予相应药物灌胃干预。HE和Masson染色分别观察肺组织肺泡炎和肺间质纤维化的程度;免疫组织化学技术检测肺组织CD31、CD34蛋白表达,RT-PCR法检测肺组织血管生长内皮因子(VEGF)、碱性成纤维细胞生长因子(bFGF)、缺氧诱导因子-1α(HIF-1α)及Ⅰ型胶原(COL-Ⅰ)、Ⅲ型胶原(COL-Ⅲ)mRNA表达。结果:天龙竭高、中、低剂量均可抑制肺间质纤维化大鼠肺部肺泡炎和肺纤维化的发生。与模型组比较,各给药组均能显著降低肺组织中CD31、CD34蛋白表达及VEGF、bFGF、HIF-1α、COL-Ⅰ、COL-ⅢmRNA表达(P<0.01,P<0.05)。结论:天龙竭可通过抑制血管重构,降低肺纤维化模型大鼠肺组织中炎症发生,减少胶原沉积,而发挥抗肺间质纤维化的作用。Objective:To investigate the mechanism of anti-interstitial lung fibrosis by observing the effects of the cloud medicine Tianlongjie on vascular remodeling in rats with bleomycin-induced interstitial lung fibrosis.Methods:Sixty Wistar rats were randomly divided into a blank group,a model group,a group with high,medium and low doses of Tianlongjie,and a group with pirfenidone,with 10 rats in each group.Except for the blank group,all groups were used to replicate the rat model of interstitial lung fibrosis by intratracheal drip bleomycin injection.HE and Masson staining were used to observe the degree of alveolitis and interstitial fibrosis,respectively.Immunohistochemical techniques were used to detect the expression of CD31 and CD34 proteins,and RT-PCR was used to detect the mRNA expression of vascular endothelial growth factor(VEGF),basic fibroblast growth factor(bFGF),hypoxia-inducible factor 1α(HIF-1α)and collagen type I and type I(COL-I,COL-I).
