摘  要:目的:研究入山虎乙酸乙酯和正丁醇部位的抗炎镇痛活性,并初步分析其活性成分。方法:以二甲苯致小鼠耳肿胀法、角叉菜胶致小鼠足趾肿胀法的炎症模型分析入山虎乙酸乙酯和正丁醇部位抗炎效果;以热板法和醋酸扭体法的疼痛模型考察入山虎的乙酸乙酯、正丁醇部位镇痛效果。采用超高效液相色谱-四极杆飞行时间串联质谱(UPLC-Q-TOF-MS/MS)技术快速鉴定入山虎两个部位的化学成分。结果:与模型组比较,乙酸乙酯部位组、正丁醇部位组小鼠耳肿胀度、足趾肿胀度显著降低(P<0.01,P<0.05),扭体次数显著减少(P<0.01),药后30 min痛阈值显著提高(P<0.01)。初步筛选得到入山虎乙酸乙酯和正丁醇化合物67个,初步鉴定结构53个,未知14个。结论:入山虎乙酸乙酯和正丁醇部位有较好的抗炎镇痛活性,推测两部位的活性成分为生物碱类成分。Objective:To examine Rushanhu's active ingredients and investigate the analgesic and anti-inflammatory properties of its ethyl acetate and n-butanol components.Methods:The analgesic effects of the ethyl acetate and n-butanol sites of Rushanhu were examined using the pain models of the hot-plate method and the cool-acid-twist method,while the anti-inflammatory effects were examined using the inflammation models of xylene-induced ear swelling method in mice and carrageenan gum-induced toe swelling method in mice.The two components of Rushanhu were quickly identified by using the ultra performance liquid chromatography-quadrupole time-of-flight tandem mass spectrometry(UPLC-Q-TOF-MS/MS)technology.Results:Compared with the model group,mice in the ethyl acetate and n-butanol groups showed significantly lower levels of ear and toe swelling(P<0.01,P<0.05),the number of twisting is significantly reduced(P<0.01),as well as a significantly higher pain threshold value 30 minutes after the drug(P<0.01).Of the 67 compounds of Rushanhu ethyl acetate and n-butanol that were found through preliminary screening,53 had structures that were tentatively characterized and 14 were unknown.Conclusion:Rushanhu's ethyl acetate and n-butanol components exhibit strong analgesic and anti-inflammatory properties,and it is thought that these compounds are alkaloids.
