紫花黄华薄层色谱鉴别方法开发研究

摘　　要：目的 建立藏药紫花黄华饮片、紫花黄华膏的薄层色谱鉴别方法。方法 以紫花黄华对照药材为对照,对薄层板类型、展开剂种类、点样量进行薄层色谱鉴别方法的考察优化,并对环境温度、湿度分别进行了方法耐用性考察,确定紫花黄华饮片最佳薄层色谱条件;采用相同方法鉴别紫花黄华膏,确定紫花黄华膏最佳薄层色谱条件。结果 采用2%NaOH碱性硅胶G薄层板,以丙酮-乙酸乙酯-甲醇(2∶1∶1,每20 mL内加2~3滴浓氨水)为展开剂,置紫外光灯(254 nm)下检视。紫花黄华饮片、紫花黄华膏供试品色谱中,在与对照药材色谱相应的位置上,显示相同颜色的主斑点。结论 新建的方法具有操作简便、高效快速以及高灵敏度的特点,可同时应用于紫花黄华饮片和紫花黄华膏,为紫花黄华提供了快速定性鉴别方法和依据。Objective To develop a thin-layer chromatography(TLC)method for the identification of Thermopsis barbata Benth and its thermopsis radix extract.Methods Using medicinal herbs of Thermopsis barbata Benth as reference,the type of sil-ica gel plates,the type of developing agent,and the sample size were meticulously optimized for TLC identification.Additional-ly,the robustness of the method was evaluated under varying temperatures and humidity conditions.To determine the best condi-tions for thermopsis radix extract,the same method was used to identify thermopsis radix extract.Results Using a silica gel G plate(2%NaOH),with acetone-ethyl acetate-methanol(2:1:1,add 2~3 drops of concentrated ammonia per 20 mL as the developing agent,and inspecting under ultraviolet(UV)lamp(254 nm).In the chromatogram of Thermopsis barbata Benth and thermopsis radix extract,spots of the same color appear at the corresponding positions of the reference medicinal material chro-matogram.Conclusion The new method is simple,fast,and highly sensitive,and can be applied to both Thermopsis barbata Benth decoction pieces and Thermopsis barbata Benth ointment providing a rapid qualitative identification method and basis for Thermopsis barbata Benth.
