摘 要:目的探讨紫檀芪(PTE)对糖尿病性白内障模型小鼠早期白内障发展的影响。方法实验分为对照组(等体积生理盐水灌胃),模型组(等体积生理盐水灌胃),PTE高、中、低剂量组(80,40,20 mg/kg灌胃)和PTE+pcDNA-TXNIP组(80 mg/kg灌胃+尾静脉一次性注射5 nmol pcDNA-TXNIP质粒),予小鼠腹腔一次性注射2%链脲佐菌素溶液(10 mg/kg)以复制糖尿病小鼠模型,6周后即得糖尿病性白内障小鼠模型。建模成功后,各组小鼠予相应药物或生理盐水,每天灌胃1次,连续14 d。检测小鼠空腹血糖值和晶状体浑浊程度;苏木素-伊红染色,观察小鼠晶状体病理形态;酶联免疫吸附法检测血清中白细胞介素6(IL-6)、白细胞介素1β(IL-1β)、肿瘤坏死因子-α(TNF-α)和晶状体中丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)水平;Western blot法检测晶状体中硫氧还蛋白互作蛋白(TXNIP)、半胱氨酸蛋白酶-1(Caspase-1)的蛋白表达水平。结果模型组小鼠晶状体肿胀,皮质纤维紊乱且有变性样坏死,可见血管渗漏、迂曲、扩张等异常;PTE高、中、低剂量组小鼠晶状体肿胀程度减轻,皮质纤维排列较模型组整齐;PTE+pcDNA-TXNIP组小鼠晶状体病理形态与模型组相似。与模型组比较,PTE高、中、低剂量组小鼠空腹血糖值和晶状体浑浊程度评分均显著降低(P<0.05),血清中IL-6、IL-1β、TNF-α及晶状体中MDA水平和TXNIP、Caspase-1蛋白表达水平均显著降低(P<0.05),晶状体中SOD、GSH-Px水平均显著升高(P<0.05)。与PTE高剂量组比较,PTE+pcDNA-TXNIP组小鼠空腹血糖值和晶状体浑浊程度评分均显著升高(P<0.05),血清中IL-6、IL-1β、TNF-α及晶状体中MDA水平和TXNIP、Caspase-1蛋白表达水平均显著升高(P<0.05),晶状体中SOD、GSH-Px水平均显著降低(P<0.05)。结论PTE可能通过下调TXNIP介导的氧化应激反应,增强糖尿病性白内障模型小鼠晶状体抗氧化酶活性,减轻氧化应激反应,延缓Objective To investigate the effect of pterostilbene on early cataract in diabetic cataract model mice.Methods The mice were randomly divided into the control group(intragastric administration of an equal volume of normal saline),the model group(intragastric administration of an equal volume of normal saline),high-,medium-,and low-dose PTE groups(intragastric administration at doses of 80,40,and 20 mg/kg PTE respectively),and a PTE+pcDNA-TXNIP group(intragastric administration at a dose of 80 mg/kg+a single tail vein injection of 5 nmol pcDNA-TXNIP plasmid).Mice were given a single intraperitoneal injection of a 2%streptozotocin solution(10 mg/kg)to establish a diabetic mouse model.After 6 weeks,a mouse model of diabetic cataract was obtained.After the successful establishment of the model,the mice in each group were given the corresponding drugs or normal saline,administered by intragastric gavage once a day for 14 consecutive days.The fasting blood glucose levels of the mice and the degree of lens opacity were detected;hematoxylin-eosin(HE)staining was used to observe the pathological morphology of the lens;the enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of interleukin-6(IL-6),interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α)in the serum and the levels of malondialdehyde(MDA),superoxide dismutase(SOD),and glutathione peroxidase(GSH-Px)in the lens tissue.Western blot method was used to detect the protein expression levels of thioredoxin-interacting protein(TXNIP)and cysteine protease-1(Caspase-1)in the lens.Results In the model group,the lenses of the mice were swollen,the cortical fibers were disordered with degenerative necrosis,and abnormalities such as vascular leakage,tortuosity,and dilation were observed.In the high-,medium-,and low-dose PTE groups,the degree of lens swelling in the mice was reduced,and the arrangement of the cortical fibers was neater compared with that in the model group.The pathological morphology of the lenses in the mice of the PTE+pcDNA-TXNIP group wa
