摘 要:目的建立百玫解郁发酵酒的质量标准。方法采用薄层色谱(TLC)法对制剂中玫瑰花、淡竹叶和甘草进行定性鉴别。采用高效液相色谱法(HPLC)测定儿茶素、葛根素、夏佛塔苷、柚皮苷和橙皮苷的含量,色谱柱为Agilent Eclipse XDB-C_(18)柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.2%磷酸水溶液(梯度洗脱),流速为1.0 mL/min,检测波长为280 nm,柱温为30℃,进样量为5μL。结果玫瑰花、淡竹叶、甘草的TLC图斑点显色清晰,分离度良好,阴性对照无干扰。儿茶素、葛根素、夏佛塔苷、柚皮苷、橙皮苷质量浓度分别在28.00~1792.13μg/mL、1.18~75.71μg/mL、2.50~159.78μg/mL、3.73~238.40μg/mL、0.78~49.76μg/mL范围内与峰面积线性关系良好(r>0.9996,n=7);精密度、稳定性、重复性试验结果的RSD均小于2.0%;平均加样回收率分别为99.62%,97.75%,98.14%,98.05%,96.99%,RSD分别为0.92%,1.42%,1.91%,1.28%,2.03%(n=6)。结论该研究所建方法操作简单便捷,结果准确可靠,可用于百玫解郁酒的质量控制。Objective To establish a quality standard for Baimei Jieyu fermented wine.Methods Thin-layer chromatography(TLC)was used for qualitative identification of Rosa rugosa,Lophatherum gracile,and Glycyrrhiza uralensis in the preparation.High-performance liquid chromatography(HPLC)was employed to determine the content of catechin,puerarin,schaftoside,naringin,and hesperidin.The chromatographic column was Agilent Eclipse XDB-C_(18) column(250 mm×4.6 mm,5μm),with a mobile phase of acetonitrile-0.2%phosphoric acid aqueous solution(gradient elution).The flow rate was 1.0 mL/min,the detection wavelength was 280 nm,the column temperature was 30℃,and the injection volume was 5μL.Results The TLC spots of Rosa rugosa,Lophatherum gracile,and Glycyrrhiza uralensis were clearly colored,well-separated,and showed no interference from the negative control.The mass concentrations of catechin,puerarin,schaftoside,naringin,and hesperidin exhibited good linear relationships with peak area within the ranges of 28.00-1,792.13μg/mL,1.18–75.71μg/mL,2.50–159.78μg/mL,3.73–238.40μg/mL,and 0.78–49.76μg/mL,respectively(r>0.9996,n=7).The RSD values for precision,stability,and repeatability tests were all less than 2.0%.The average spiked recovery rates were 99.62%,97.75%,98.14%,98.05%,and 96.99%,with RSD values of 0.92%,1.42%,1.91%,1.28%,and 2.03%,respectively(n=6).Conclusion The established method is simple,convenient,accurate,and reliable,making it suitable for quality control of BaiMei JieYu wine.
