摘 要:骨质疏松症(Osteoporosis,OP)是一种以骨量降低和骨组织微结构破坏为特征的代谢性骨病。牛初乳碱性蛋白(Colostrum basic protein,CBP)是一种分子质量为1~30 ku的活性蛋白,它以牛初乳为原料,经杀菌、脱脂、离心分离去除其余蛋白,再经微滤、超滤和冷冻干燥等工艺制成。CBP具有促进成骨细胞的分化矿化,进而缓解OP发病进程的潜能。文章旨在探究以乳矿物盐、维生素D、CBP三者联合而成的CBP组合物对糖皮质激素诱导的小鼠骨质疏松症(Glucocorticoid-induced osteoporosis,GIOP)的改善作用及其可能的作用途径。研究利用糖皮质激素药物地塞米松构建GIOP小鼠模型,探究CBP、乳矿物盐、维生素D组成的CBP组合物对GIOP的改善作用。结果表明,CBP及其组合物能够显著改善GIOP小鼠体质量(P<0.05)、骨小梁数(P<0.05)、骨小梁分离度(P<0.01),显著降低股骨骨骺区破骨细胞阳性数量(P<0.01)以及显著改善骨钙素水平(P<0.05)。以上研究表明,CBP组合物能够提高成骨细胞分化程度,平衡成骨细胞和破骨细胞,有效改善GIOP小鼠骨质疏松症状,并为富含CBP的乳制品的科学利用和产品开发提供理论依据。Osteoporosis(OP)is a metabolic bone disease characterized by reduced bone mass and microstructural destruction of bone tissue.Colostrum basic protein(CBP)is a molecular mass of about 1–30 ku active protein,it is made from bovine colostrum by sterilization,degreasing,centrifuge,and then by microfiltration,ultrafiltration and freeze-drying.CBP can promote the differentiation and mineralization of osteoblasts and alleviate the pathogenesis of OP.This article aims to explore the improving effect of CBP composition combined with milk mineral salt,vitamin D and CBP on glucocorticoid-induced osteoporosis(GIOP)in mice and its possible action pathways.The study first used the glucocorticoid drug dexamethasone to construct a GIOP mouse model to explore the improvement effect of a CBP composition composed of CBP,milk mineral salts,and vitamin D on osteoporosis.The results showed that CBP and its compositions could significantly improve the body weight(P<0.05),trabecular bone number(P<0.05),trabecular bone separation(P<0.01),significantly reduce the number of osteoclasts positive in epiphyseal region of femur(P<0.01),and improve the level of osteocalcin in GIOP mice(P<0.05).The above studies have shown that the CBP composition can increase osteoblast differentiation activity,balance osteoblasts and osteoclasts,effectively improve osteoporosis symptoms in GIOP mice,and provide a theoretical basis for the scientific utilization and product development of CBP-rich dairy products.
