枸杞多糖抑制NLRP3炎症小体通路保护肾缺血再灌注损伤的作用

摘  要:目的探讨核苷酸结合寡聚化结构域样受体蛋白3[(nucleotide-binding oligomerization domain,NOD)-like receptor protein 3,NLRP3]炎症小体通路的活性在枸杞多糖(Lycium barbarum polysaccharide,LBP)改善肾缺血再灌注损伤的作用,为防治肾缺血再灌注损伤提供依据。方法于2023年1月—2023年12月,制造大鼠肾缺血再灌注模型,随机分为假手术组、模型组和LBP治疗组。采用ELISA分析大鼠血清中TNF-α、白介素-1β(interleukin-1β,IL-1β)和白介素-18(interleukin-18,IL-18)含量;采用免疫印迹法检测大鼠肾组织中NLR3炎症小体通路蛋白:NLRP3、凋亡相关的斑点样蛋白(apoptosis-associated speck-like protein,ASC)、天冬氨酸半胱氨酸蛋白酶-1(cysteinyl aspartate specific proteinase-1,Caspase-1)和IL-1β的含量。2组数据间的比较采用t检验,3组数据间的比较采用单因素方差分析。结果与假手术组比较,模型组大鼠肾组织坏死严重,血液中肌酐和尿素氮含量明显升高(t=14.463、18.727,P<0.01),血清中TNF-α、IL-1β、IL-18含量均升高(t=9.875、11.335、7.669,均P<0.01),组织中NLRP3炎症小体通路蛋白:NLRP3、ASC、Caspase-1和IL-1β含量均升高(t=11.968、13.292、16.245、15.154,均P<0.01)。与模型组比较,LBP治疗组大鼠肾组织坏死减轻,血液中肌酐和尿素氮含量明显降低(t=10.957、14.125,P<0.01),血清中TNF-α、IL-1β、IL-18含量均降低(t=6.464、7.553、5.026,均P<0.01),组织中NLRP3炎症小体通路蛋白:NLRP3、ASC、Caspase-1和IL-1β含量均降低(t=6.866、8.592、9.692、9.805,均P<0.01)。结论枸杞多糖能改善缺血再灌注引起的肾组织损伤,与其抑制NLRP3炎症小体通路的活性有关。Objective Exploring the role of nucleotide binding oligomerization domain-like receptor protein 3(NLRP3)inflammasome pathway activity in the improvement of renal ischemia-reperfusion injury by Lyciumbar barum polysaccharide(LBP),to provide a basis for the prevention and treatment of renal ischemia-reperfusion injury.Methods From January 2023 to December 2023,a rat renal ischemia-reperfusion model was manufactured and randomly divided into a sham surgery group,a model group,and an LBP treatment group.ELISA was used to analyze the levels of TNF-α,interleukin-1β(IL-1β),and interleukin-18(IL-18)in rat serum.Immunoblotting was used to detect the content of NLR3 inflammasome pathway proteins,including NLRP3,apoptosis-associated spotted-like protein(ASC),cysteinyl aspartate specific proteinase-1(Caspase-1),and IL-1βin rat kidney tissues.The comparison between two sets of data was conducted using the t-test,while the comparison among three sets of data was conducted using one-way analysis of variance.Results Compared with the sham surgery group the renal tissue of the model group rats showed severe necrosis,with significantly increased content of creatinine and urea nitrogen in the blood(t=14.463,18.727,all P<0.01),elevated content of TNF-α,IL-1β,and IL-18 in the serum(t=9.875,11.335,7.669,all P<0.01),and elevated content of NLRP3 inflammasome pathway proteins:NLRP3,ASC,Caspase-1,and IL-1βin the tissues(t=11.968,13.292,16.245,15.154,all P<0.01).Compared with the model group,the LBP treatment group rats showed reduced renal tissue necrosis,creatinine and urea nitrogen content in the blood were significantly decreased(t=10.957,14.125,all P<0.01),TNF-α,IL-1β,and IL-18 content in the serum were decreased(t=6.464,7.553,5.026,all P<0.01),and NLRP3 inflammasome pathway proteins:NLRP3,ASC,Caspase-1,and IL-1βcontent in the tissue were decreased(t=6.866,8.592,9.692,9.805,all P<0.01).Conclusion Lycium barbarum polysaccharides can improve renal tissue injury caused by ischemia-reperfusion,which is related to their inhibiti
