合苞橐吾指纹图谱的建立及含量测定

摘  要:目的:建立合苞橐吾的HPLC指纹图谱,并对4个成分进行含量测定。方法:采用“中药色谱指纹图谱相似度评价系统(2004 A版)”建立10批合苞橐吾的HPLC指纹图谱,并进行相似度评价,采用SPSS 23.0、SIMCA 14.1软件进行聚类分析、主成分分析、正交偏最小二乘判别分析,以变量重要性投影(VIP)值大于1为标准筛选影响合苞橐吾质量的标志物;测定成分的含量。结果:10批合苞橐吾指纹图谱共有20个共有峰,相似度为0.845~0.999;指认了4个成分,分别为绿原酸、芦丁、木犀草苷、异绿原酸A,上述4个成分的含量分别为17.42~36.62 mg/g、0.99~1.81 mg/g、0.36~0.67 mg/g、1.03~2.17 mg/g。结论:建立的合苞橐吾HPLC指纹图谱及成分含量测定方法操作简便,结果准确,可用于该药材的质量评价。Objective:To establish HPLC fingerprint of Ligularia schmidtii leaves and to determine the contents of 4 components.Methods:HPLC fingerprint of 10 batches of Ligularia schmidtii leaves was established by"Traditional Chinese medicine chromatographic fingerprint similarity evaluation system(2004 A edition)",and similarity evaluation was carried out.SPSS 23.0 and SIMCA 14.1 software were used for cluster analysis,principal component analysis and orthogonal partial least squares discriminant analysis.With the variable importance projection(VIP)value greater than 1 as the standard,the markers affecting the quality of Ligularia schmidtii leaves were screened,the content of the compounds were determined.Results:There were 20 common peaks in the fingerprints of 10 batches of Ligularia schmidtii leaves,and the similarity was 0.845 to 0.999.Four components were identified,namely chlorogenic acid,rutin,luteolin and isochlorogenic acid A,the contents of which were 17.42~36.62 mg/g,0.99~1.81 mg/g,0.36~0.67 mg/g and 1.03~2.17 mg/g,respectively.Conclusion:The established HPLC fingerprint and component content determination method of Ligularia schmidtii leaves is simple and accurate,and can be used for the quality evaluation of Ligularia schmidtii.
