沙枣果总黄酮对大鼠PCOS-IR模型的干预作用研究

摘  要:目的探究沙枣果总黄酮对多囊卵巢综合征合并胰岛素抵抗(PCOS-IR)大鼠模型的干预治疗作用。方法采用绒毛膜促性腺激素(HCG)+胰岛素(INS)+高脂高糖饲料方法建立大鼠PCOS-IR模型,同时设立空白组。将造模成功的大鼠随机分为5组:模型组(等体积生理盐水)、二甲双胍组(104 mg/kg)、低剂量黄酮组(50 mg/kg)、高剂量黄酮组(200 mg/kg)和二甲双胍+高剂量黄酮组(104 mg/kg+200 mg/kg),每组5只,每日给予相应药物灌胃1次,空白组与模型组每日给予等体积生理盐水,连续给药26 d后称重取材,检测相关指标。结果与模型组相比,各治疗组大鼠卵巢组织结构紊乱、黄体减少、囊性卵泡和闭锁卵泡数目增多的病理损伤均有不同程度改善;同时各治疗组体重、子宫指数(除了低剂量黄酮组)、卵巢指数、空腹胰岛素(FINS)、HOMA-IR、睾酮(T)、LH均有不同程度的显著下降(P<0.05),雌二醇(E_(2))(除了低剂量黄酮组)、孕酮(P)、FSH水平则有不同程度的显著上升(P<0.05),以二甲双胍+高剂量黄酮组变化最为显著(P<0.01);低剂量黄酮组与模型组相比,子宫指数呈现出一定下降趋势,E_(2)水平呈现一定上升趋势,但差异尚无统计学意义(P>0.05);与高剂量黄酮组相比,二甲双胍+高剂量黄酮组的体重、HOMA-IR、LH水平均显著下降(P<0.05),E_(2)、P、FSH水平均显著升高(P<0.05),T、子宫指数、卵巢指数也有一定改变但是均尚无统计学意义(P>0.05)。结论沙枣果总黄酮对PCOS-IR大鼠模型具有一定干预治疗作用,本研究可为沙枣果总黄酮治疗PCOS-IR提供初步实验理论依据。Objective:To investigate the therapeutic effect of total flavone of oleaster fruit on the rat model of PCOS complicated with insulin resistance(PCOS-IR).Methods:The PCOS-IR model was established by HCG+insulin+high-fat and high-sugar diet.A blank group was established simultaneously.The rats with successful modeling were randomly divided into five groups:model group,metformin group(104 mg/kg),low dose flavone group(50 mg/kg),high dose flavone group(200 mg/kg)and metformin combined with high dose flavone group(104 mg/kg+200 mg/kg),with five rats in each group.Rats in the blank and the model group received equal volume normal saline.After continuous administration for 26 days,the samples were weighed and the relevant indexes were detected.Results:Compared with the model group,the pathological damages of ovarian tissue structure disorder,corpus luteum reduction,and the increase of the number of cystic follicles and atretic follicles improved with different degrees.At the same time,body mass,uterine index(except for low dose flavone group),ovarian index,fasting insulin level,the homeostatic model assessment for insulin resistance(HOMA-IR),testosterone(T)and LH decreased to varying degrees(P<0.05),while estradiol(E_(2),except for low dose flavone group),progesterone(P)and FSH levels increased(P<0.05),especially in metformin combined with high dose flavone group(P<0.01).Although the uterine index showed a certain downward trend and the E_(2)level showed an upward trend in low dose flavone group,the differences were not statistically significant when compared with the model group(P>0.05).Compared with high dose flavone group,there was a significant decrease in body mass,HOMA-IR and LH level(P<0.05),and a significant increase in the levels of E_(2),P and FSH in metformin combined with high dose flavone group(P<0.05).The levels of T,uterine index and ovarian index also changed,but the differences were not statistically significant(P>0.05).Conclusions:Total flavone of oleaster fruit had a certain therapeutic effect on PCOS
