摘  要:目的:应用网络药理学探讨聚精活血汤治疗弱精子症的作用机制,为其临床应用提供新的研究思路。方法:通过网络药理学数据库TCMSP、PubChem、SwissTargetPrediction获得聚精活血汤组成药物的活性成分及靶点;由GeneCards、DisGeNET、OMIM数据库中获得弱精子症相关靶点,将二者靶点取交集后获得共有靶点;利用DAVID数据库对共有靶点进行基因本体(GO)和京都基因与基因组百科全书(KEGG)富集分析;应用STRING在线数据库构建共有靶点蛋白互作网络(PPI)分析。结果:共筛选出201个聚精活血汤主要活性成分,包括谷甾醇(sitosterol)、豆甾醇(stigmasterol)、β-谷甾醇(beta-sitosterol)、木犀草素(luteolin)、丝氨酸蛋白酶等482个药物靶基因,726个疾病靶基因,取交集后得到65个共有靶基因,核心靶基因为TP53、IL6、TNF、CASP3、IL1B、MYC等;KEGG通路富集分析得到115个条目,主要为癌症通路、IL-17信号通路、PI3K-Akt信号通路等。结论:聚精活血汤通过多成分、多靶点、多通路协同调控并治疗弱精子症,为临床研究及应用提供依据。Objective:To explore the mechanism of action of Jujing Huoxue Decoction in the treatment of asthenozoospermia using network pharmacology,and provide new research ideas for its clinical application.Methods:The active ingredients and targets of Jujing Huoxue Decoction were obtained through network pharmacology databases such as TCMSP,PubChem,and SwissTargetPrediction.The targets related to asthenozoospermia were obtained from GeneCards,DisGeNET,and OMIM databases,and the common targets were obtained after the intersection of the above two targets.Gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)were enriched for common targets using the DAVID database.Common target protein interaction Network(PPI)analysis was constructed using the STRING database.Results:A total of 201 main active ingredients of Jujing Huoxue Decoction were screened,including sitosterol,stigmasterol,beta-sitosterol,luteolin,CLR,etc.There were 482 drug-target genes and 726 disease-target genes.65 common target genes were obtained after the intersection,and the core target genes were TP53,IL6,TNF,CASP3,IL1B,MYC,etc.KEGG pathway enrichment analysis obtained 115 items,mainly including the cancer pathway,IL-17 signaling pathway,and PI3K Akt signaling pathway.Conclusion:Jujing Huoxue Decoction can treat asthenozoospermia by coordinating regulation through multi-components,multi-targets,and multi-pathways,which providing a basis for clinical research and application.
