灰树花多糖对对乙酰氨基酚诱导的小鼠肝损伤的保护作用

摘  要:目的:探讨灰树花多糖(GFP)对对乙酰氨基酚(APAP)诱导的小鼠肝损伤的保护作用。方法:选择健康成年清洁级BALB/c雄性小鼠40只,随机分为对照组、模型组和低、高剂量组,每组10只。低、高剂量组分别灌胃72 mg·kg^(-1)、144 mg·kg^(-1)灰树花多糖,连续10 d,对照组和模型组灌胃等量生理盐水。除对照组腹腔注射生理盐水外,其余各组均腹腔注射对乙酰氨基酚溶液(500 mg·kg^(-1))造模,24 h后取材观察各组小鼠肝脏外观形态,计算肝脏指数,检测血浆ALT和AST水平、肝匀浆还原型GSH水平,HE染色观察肝脏组织形态结构。结果:模型组肝脏颜色变暗,表面不平整、有肉眼可见的暗红色出血灶,质地发硬;低、高剂量组肝脏外观相对接近于正常;低、高剂量组肝脏指数、血浆AST和ALT水平显著低于模型组(P<0.05);高剂量组肝脏组织GSH水平显著高于模型组(P<0.001);模型组镜下观察肝索排列紊乱,肝细胞肿胀并脂肪变性,内充满脂滴,核固缩、碎裂,肝血窦淤血狭窄、消失;低剂量组肝脏组织损伤有所减轻,肝细胞内脂滴减少、肿胀减轻,肝索及肝血窦可见;高剂量组肝脏组织接近于正常。结论:灰树花多糖能够提高肝组织还原型GSH水平,抑制膜脂质过氧化,提高肝细胞氧化耐受性进而稳定肝细胞膜,有效防治过量APAP引起的急性肝损伤。Objective:To explore the protective effect of Grifola frondosa polysaccharide(GFP)on acetaminophen(APAP)-induced liver injury in mice.Methods:Forty healthy adult clean-grade BALB/c male mice were selected and randomly divided into the control group,the model group,and the low-and the high-dose groups,with 10 mice in each group.The low-and high-dose groups were administrated with 72 mg·kg^(-1)and 144 mg·kg^(-1)Grifola frondosa polysaccharide respectively for 10 consecutive days;the control group and the model group were administrated with normal saline.Except for the control group,which was injected intraperitoneally with normal saline,all other groups were intraperitoneally injected with acetaminophen solution(500 mg·kg^(-1))to create a model.After 24 hours,the livers of the mice in each group were taken to observe the appearance and morphology,the liver index was calculated,the plasma ALT and AST levels and liver homogenate reduced GSH levels were detected,and the liver tissues were stained by HE to observe the morphology and structure.Results:The liver in the model group became darker in color,with an uneven surface,dark red bleeding lesions visible to the naked eye,and a hard texture.The liver appearance in the low-and high-dose groups was relatively close to normal.The liver index,plasma AST and ALT levels in the low-and high-dose groups were significantly lower than those in the model group(P<0.05).The GSH level in the liver tissue of the high-dose group was significantly higher than that of the model group(P<0.001).In the model group,it was observed under the microscope that the arrangement of the liver cords was disordered,the liver cells were swollen and fatty,filled with lipid droplets,the nuclei were pyknotic and fragmented,and the liver sinusoids were congested,narrowed,and disappeared;in the low-dose group,liver tissue damage was reduced,lipid droplets and swelling in liver cells were reduced,and liver cords and liver sinusoids were visible;liver tissue in the high-dose group was close to normal.Co
