摘 要:目的:建立超高效液相色谱-三重四级杆质谱联用(UPLC-QQQ-MS/MS)技术测定防己黄芪汤中18种成分含量的测定方法,为科学评价防己黄芪汤的质量提供依据。方法:采用UPLC-QQQ-MS/MS技术建立对防己黄芪汤中多成分同时测定的分析方法,对复方进行多成分表征。结果:建立了灵敏度高、分析质量可靠、分析效率高且能对防己黄芪汤复方中多成分同时测定的UPLC-QQQ-MS/MS分析方法,并应用于防己黄芪汤中的生物碱类(木兰花碱、防己诺林碱、粉防己碱)、皂苷类(黄芪皂苷Ⅲ、黄芪甲苷、黄芪皂苷Ⅰ、甘草次酸)、黄酮类(毛蕊异黄酮葡萄糖苷、芒柄花苷、5-O-甲基维斯阿米醇苷、毛蕊异黄酮、芒柄花素、甘草苷、异甘草苷、甘草素、异甘草素)和萜类(白术内酯Ⅲ、白术内酯Ⅱ)共18种成分的定量分析研究,其中粉防己碱为防己黄芪汤复方中含量最高的生物碱。结论:该方法专属性强,可用于防己黄芪汤中多种成分的定量检测。Objective:To establish a method for determining eighteen components in Fangji Huangqi Decoction(FJHQD)by UPLCQQQ-MS/MS,in order to provide a basis for the scientific evaluation of the quality of FJHQD.Methods:A method for simultaneous determination of multi-components in FJHQD by UPLC-QQQ-MS/MS technology was established to characterize the multi-components of the compound.Results:A UPLC-QQQ-MS/MS analysis method for simultaneous determination of multi-components in FJHQD,with high sensitivity,reliable analysis quality,and high efficiency,was established.This method was applied to study the quantitative analysis of 18 components in FJHQD,including alkaloids(magnoflorine,fangchinoline,tetrandrine),saponins(astragalosideⅢ,astragalosideⅣ,astragalosideⅠ,glycyrrhetic acid),flavonoids(calycosin-7-glucoside,ononin,5-O-methylvisammioside,calycosin,formononetin,liquiritin,liquiritigenin,neoisoliquritin,isoliquiritigenin),and terpenes(atractylenolideⅢ,atractylenolideⅡ).Additionally,tetrandrine was the alkaloid with the highest content in FJHQD compound.Conclusion:This method is specific and can be used for the quantitative determination of various components in FJHQD.
