止痒促愈乳膏对特应性皮炎小鼠模型的治疗作用及其炎症相关机制的研究

摘  要:目的:观察止痒促愈乳膏对2,4-二硝基氯苯(DNCB)诱导的特应性皮炎(AD)小鼠模型的治疗作用,并探讨其潜在机制。方法:取Balb/c小鼠32只,随机分为止痒促愈乳膏组、乳膏基质组、模型组和空白对照组,除空白对照组外,其余各组小鼠背部涂抹DNCB诱导建立AD模型,止痒促愈乳膏组、乳膏基质组分别使用相对应的药物治疗,治疗14 d后观察其皮损、搔抓次数;苏木精-伊红(H-E)染色观察皮肤病理变化、表皮厚度;酶联免疫吸附试验(ELISA)法检测血清免疫球蛋白E(IgE)水平;蛋白免疫印迹(Western blot)法检测皮肤组织中磷酸化-Janus激酶(p‑JAK)/JAK1、JAK、信号转导和转录激活(STAT)、p-STAT3的表达变化。结果:与空白对照组相比,模型组小鼠的皮损评分、搔抓次数、表皮厚度、血清IgE水平、JAK1/STAT3通路相关蛋白表达上升(P<0.05);与模型组相比,止痒促愈乳膏组小鼠的皮损评分、搔抓次数、表皮厚度、血清IgE水平、JAK1/STAT3通路相关蛋白表达下降(P<0.05);与乳膏基质组相比,止痒促愈乳膏组小鼠的皮损评分、搔抓次数、表皮厚度、JAK1/STAT3通路相关蛋白表达下降(P<0.05)。结论:止痒促愈乳膏可以改善AD小鼠模型的皮损、搔抓次数、皮肤病理、血清IgE水平,通过抑制JAK1/STAT3通路激活,改善AD小鼠的炎症反应。Objective:To observe the therapeutic effect of Zhi-Yang-Cu-Yu Cream on atopic dermatitis(AD)mouse model induced by 2,4-dinitrochlorobenzene(DNCB)and explore its potential mechanism.Methods:32 Balb/c mice were randomly divided into Zhi-YangCu-Yu Cream group,cream base group,model group and blank control group.Except for the blank control group,DNCB was applied on the back of the mice in the other groups to induce AD models.The Zhi-Yang-Cu-Yu Cream group and the cream base group were treated with corresponding drugs respectively.After 14 days′treatment,the skin lesions and scratching times were observed.Skin pathological changes and epidermal thickness were observed by hematoxylin-eosin(H-E)staining.Serum immunoglobulin E(IgE)level was detected by enzyme-linked immunosorbent assay(ELISA).The level changes of phosphorylated-Janus kinase(p-JAK)/JAK1,JAK,signal transduction and activation of transcription(STAT)and p-STAT3 in skin tissue were detected by Western blot.Results:Compared with the blank control group,the skin lesion score,scratching times,epidermal thickness,serum IgE level and the expression of JAK1/STAT3 pathway-related proteins were increased in the model group(P<0.05).Compared with the model group,the skin lesion score,scratching times,epidermal thickness,serum IgE level and the expression of JAK1/STAT3 pathway-related protein were decreased in the Zhi-Yang Cu-Yu Cream group(P<0.05).Compared with the cream base group,the skin lesion score,scratching times,epidermal thickness and the expression of JAK1/STAT3 pathway-related proteins were decreased in the Zhi-Yang-Cu-Yu Cream group(P<0.05).Conclusion:ZhiYang-Cu-Yu Cream can improve skin lesion,scratch frequency,skin pathology and serum IgE level of AD mouse model,and improve the inflammatory response of AD mouse by inhibiting the activation of JAK1/STAT3 pathway.
