摘 要:目的 探讨微小核糖核酸-15a(miRNA-15a)对高糖诱导的腹膜间皮细胞上皮间质转化的影响。方法 将传代培养的HPMCs分为低糖对照组(葡萄糖浓度25mmol/L)、中糖组(葡萄糖浓度75mmol/L)和高糖组(葡萄糖浓度125mmol/L),使用实时荧光定量PCR(qRT-PCR)与Western印记技术(Western blot),检测miR-15a、VEGFmRNA和VEGF蛋白的表达水平。继续将HPMCs置于高糖培养基中(葡萄糖浓度125mmol/L),进行分组培养,分为:高糖对照组、高糖+control组、高糖+mimics组、高糖+inhibitor组,检测miR-15a和VEGF mRNA的表达,以及纤维化相关蛋白VEGF、Col-IV、FN1、α-SMA、E-cadherin蛋白表达。结果 高糖环境使miR-15a表达下调,VEGF mRNA表达水平上调,呈浓度依赖性,与VEGF蛋白表达结果一致(P<0.05)。高糖条件下,上调miR-15a的表达,VEGF mRNA的表达减少,VEGF、Col-IV、FN1、α-SMA蛋白减少,E-cadherin蛋白表达增加,抑制miR-15a的作用后VEGF mRNA的表达增加,VEGF、Col-IV、FN1、α-SMA蛋白表达增加,E-cadherin蛋白表达减少(P<0.05)。结论 miRNA-15a参与并抑制高糖诱导的人腹膜间皮细胞(HPMCs)上皮间质转化。Objective To investigate the effect of microRNA-15a(miRNA-15a)on high glucose-induced epithelial-mesenchymal transition(EMT)in human peritoneal mesothelial cells(HPMCs).Methods Passaged HPMCs were divided into three groups:low-glucose control group(glucose concentration 25 mmol/L),medium-glucose group(glucose concentration 75 mmol/L),and high-glucose group(glucose concentration 125 mmol/L).Real-time quantitative PCR(qRT-PCR)and Western blotting were used to measure the expression levels of miRNA-15a,VEGF mRNA,and VEGF protein.HPMCs were then cultured in high-glucose medium(glucose concentration 125 mmol/L)and further divided into four subgroups:high-glucose control group,high-glucose+control group,high-glucose+mimics group,and high-glucose+inhibitor group.The expression of miRNA-15a and VEGF mRNA,as well as the fibrosis-related proteins VEGF,Col-IV,FN1,α-SMA,and E-cadherin,were assessed.Results High glucose conditions led to downregulation of miRNA-15a expression and upregulation of VEGF mRNA expression in a concentration-dependent manner,which was consistent with the results for VEGF protein expression(P<0.05).Under high glucose conditions,overexpression of miRNA-15a resulted in decreased VEGF mRNA expression,reduced levels of VEGF,Col-IV,FN1,andα-SMA proteins,and increased expression of E-cadherin.Inhibition of miRNA-15a led to an increase in VEGF mRNA expression,higher protein levels of VEGF,Col-IV,FN1,andα-SMA,and reduced expression of E-cadherin(P<0.05).Conclusion miRNA-15a participates in and inhibits high glucose-induced epithelial-mesenchymal transition in human peritoneal mesothelial cells.
