摘 要:本文对推荐性国家标准GB/T 5537-2008《粮油检验磷脂含量的测定》中的钼蓝比色法应用于中药膏方中磷脂含量测定进行方法学研究,并对院内制做的中药膏方进行测定,以了解中药膏方中的磷脂含量。结果:在标准曲线的制作过程中,磷在反应中浓度控制约在0.003 mg/ml,硫酸(H_(2)SO_(4))浓度约在0.53 mol/L,钼酸钠(Na_(2)MoO_(4))与硫酸联胺(N_(2)H_(6)SO_(4))的摩尔比约为40∶1即可以较好屏蔽二者反应产生的钼蓝对磷含量检测的干扰;钼酸钠在磷钼蓝反应体系中,其浓度对反应结果有明显的影响,其浓度在配制溶液浓度的约十分之一为好(即0.01034 mol/L);硫酸联氨浓度对磷钼蓝反应有较大的影响,硫酸联氨用量约为配制工作液浓度的0.45倍(即0.0005 mol/L上下为好)。在这个实验方法下检测中药膏方中的磷脂含量,测得本院10个膏方中的磷脂含量范围在29.37~61.82 mg/g之间。该研究表明GB/T 5537-2008中的磷钼蓝比色法适用于中药膏方中磷脂含量的测定。This article conducts methodological research on the application of the molybdenum blue colorimetric method in the recommended national standard"Inspection of grain and oils-Determination of phosphatide content"(GB/T 5537-2008)for the determination of phospholipid content in Chinese herbal paste,and determines the phospholipid content in Chinese herbal paste in hospital preparations to understand the content of phospholipids in the paste.The results showed that in the production process of the standard curve,the concentration of phosphorus in the reaction was controlled within a certain range of 0.003 mg/ml,the concentration of H_(2)SO_(4)was around 0.53 mmol/L,and the molar ratio of sodium molybdate to hydrazine sulfate was around 40∶1,which could effectively shield the interference of molybdenum blue generated by the reaction between Na_(2)MoO_(4)and N_(2)H_(6)SO_(4)on the detection of phosphorus content;The concentration of sodium molybdate in the phosphomolybdic blue reactant has a significant impact on the reaction results,and the optimal concentration is around one tenth of the prepared solution concentration(i.e.above 0.01034 mol/L);The concentration of hydrazine sulfate has a significant impact on the reaction of phosphorus molybdenum blue.The amount of hydrazine sulfate used is about 0.45 times the concentration of the prepared working solution(i.e.around 0.0005 mol/L is better).Using this experimental meth⁃od to detect the phospholipid content in Chinese herbal paste,the phospholipid content range of 10 formulas in our hospital was found to be between 29.37-61.82 mg/g.This study indicates that the molybdenum blue colorimetric method in GB/T 5537-2008 is suitable for the determination of phospholipid content in Chinese herbal paste.
