摘 要:近年来,众多研究揭示了中性粒细胞释放的胞外诱捕网(NET)在系统性红斑狼疮(SLE)和狼疮性肾炎(LN)的发生发展中扮演了关键角色。在抵御病原体时,中性粒细胞可释放含有解聚的染色质、组蛋白及多种抗菌成分的NET结构,以捕获并降解病原体。然而,多种NET相关蛋白可作为自身抗原诱导自身抗体的产生。NET的产生和清除失衡,可使释放的染色质、组蛋白等多种物质长期暴露,从而打破机体自我耐受机制,促进自身免疫性疾病的发生。本文将对NET在SLE及LN发病机制中的作用以及潜在的治疗方法进行综述。In recent years,accumulating evidence suggests that the neutrophil extracellular traps(NET)play a critical role in the development and progression of systemic lupus erythematosus(SLE)and lupus nephritis(LN).Upon interaction with invading microbes,neutrophils release their chromatin material,histones,as well as active lytic enzymes into the extracellular space which known as NET.NET trap the invading pathogens and degrade them with NET-associated proteolytic enzymes.However,many of these NET-associated proteins could be a source of autoantigens for autoantibody production.The imbalance between NET formation and degradation could lead to the accumulation of NET-associated proteins and prolonged exposure of NET autoantigens in the circulation.This could potentially break self-tolerance and trigger the development of autoimmune disease.This article reviews the role of NET in the pathogenesis of SLE and LN,as well as potential treatments.
