摘 要:为优化骆驼蒿总生物碱的提取工艺,以骆驼蒿总生物碱提取率为考察指标,对阳离子交换树脂提取、酸溶碱沉提取和有机溶剂提取等方法进行比较,确定其最佳提取方法为酸溶碱沉提取法。采用单因素试验结合响应面法优化酸溶碱沉提取工艺,单因素试验中考察了粉末粒度、浸泡时间、盐酸浓度及盐酸用量等影响因素。以每一单因素最佳值为中心点,采用Box-Behnken的中心组合法设计响应面的因素及水平,优化其提取工艺条件。结果显示,当粉末粒度60目、浸泡时间49 h、盐酸浓度5%、盐酸用量40 mL时,骆驼蒿总生物碱提取效果最佳,其得率为32.91%,与模型预测值33.082%接近。通过该实验研究,得到一个能较好地预测实验结果的二次模型方程,为后续对骆驼蒿总生物碱的药理作用研究打下了前期基础。This study aims to optimize the extraction process of total alkaloids from Peganum nigellastrum Bunge,taking the extraction rate of total alkaloids from Peganum nigellastrum Bunge as the evaluation index,the methods of cation exchange resin extraction,acid-solution and alkali-isolation extraction,and organic solvent extraction were compared,and the optimal extraction method was determined to be acid-solution and alkali-isolation extraction.Single factor experiments combined with response surface method were used to optimize the acid-solution and alkali-isolation extraction process.In the single factor experiment,factors such as powder particle size,soaking time,hydrochloric acid concentration,and hydrochloric acid dosage were investigated.With the optimal value of each single factor as the central point,Box-Behnken’s central combination method was used to design the factors and levels of the response surface,and optimize its extraction process conditions.The experimental results showed that when the powder particle size,soaking time,HCl concentration and HCl dosage were 60 mesh,49 h,5%and 40 mL,respectively,the extraction effect of total alkaloids from Peganum nigellastrum Bunge was the best,with a yield of 32.91%,which was close to the predicted value of the model(33.082%).Through this experimental study,a quadratic model equation was obtained that could better predict the experimental results,which laid a preliminary foundation for subsequent research on the pharmacological effects of total alkaloids from Peganum nigellastrum Bunge.
