摘  要:羧基聚苯乙烯胶乳微球是体外诊断试剂的一种,微球粒径以及与抗体的偶联能力是羧基胶乳微球的核心性能指标,建立一种简单有效、响应明显的粒径控制方法是羧基胶乳微球生产制备的关键。本文通过研究乳化剂、羧基单体用量、电解质的种类与用量,以及反应时间等因素对羧基胶乳微球粒径和形貌的影响,优化了胶乳微球制备工艺,建立了一种乳液聚合制备羧基胶乳微球粒径控制的方法。并对制得的胶乳微球进行抗体适配性表征与评价。结果表明,以十二烷基硫酸钠(SDS)为乳化剂、苯乙烯(St)为单体、丙烯酸(AA)为羧基单体(V_(St)∶V_(AA)=10∶1),K_(2)CO_(3)为电解质,可制得单分散羧基聚苯乙烯胶乳微球,电解质K_(2)CO_(3)的用量与微球粒径大小存在显著的关联性,通过在一定浓度内改变电解质K_(2)CO_(3)的用量,能够实现羧基微球在100~200 nm之间的粒径可控制备。微球偶联抗体后表现出极佳的CRP抗原响应性,吸光度达到0.469,批次重复性良好。The carboxylated polystyrene latex microsphere is a kind of in vitro diagnostic reagent.The particle size and coupling ability with antibodies are the core performance indicators of carboxyl latex microspheres.Establishing a simple,effective,and responsive particle size control method is the key to the production and preparation of carboxyl latex microspheres.In this paper,the effects of emulsifier,amount of carboxyl monomer,type and amount of electrolyte,reaction time and other factors on the particle sizes and morphologies of carboxyl latex microspheres were studied,the preparation processes of latex microspheres were optimized,and a method for controlling the particle sizes of carboxyl latex microspheres prepared by the emulsion polymerization was established.And the antibody compatibility of the prepared latex microspheres was characterized and evaluated.The results indicate that the monodisperse carboxyl polystyrene latex microspheres could be prepared by using sodium dodecyl sulfate(SDS)as an emulsifier,styrene(St)as a monomer,acrylic acid(AA)as a carboxyl monomer(VSt∶VAA=10∶1),and K_(2)CO_(3) as an electrolyte.There is a significant correlation between the dosages of electrolyte K_(2)CO_(3) and the particle sizes of the microspheres.By changing the dosages of electrolyte K_(2)CO_(3) within a certain concentration,the particle sizes of carboxyl microspheres could be controlled between 100-200 nm.The microspheres coupled with antibodies show excellent CRP antigen responsiveness,with an absorbance of 0.469 and good batch reproducibility.
