摘 要:为研究不同流动条件下,微血管栓塞的形成、演变及其对血流分布的影响,本研究基于默里定律设计并制备了一种微血管芯片。首先,利用微流控技术模拟体内动态血流环境;其次,结合激光散斑衬比成像技术实时获取流场分布;最后,分析微血管栓塞过程中栓子的动态行为及其对血流分布的影响,结合血流动力学仿真结果,揭示血流动力学因素对栓塞形成的作用。结果表明:微血管分叉顶点处由于血流速度变化,壁面剪切应力降低,容易促成栓子的聚集与生长;栓子达到临界大小后可能会脱落,进一步在下游分支引发机械堵塞;虽然侧支循环在不完全阻塞的情况下可保持微血管网络出口处流速稳定,但侧支血管中流速的增加仍可能导致血管壁的损伤,影响局部组织的血液交换效率。本研究为研究微血管栓塞机制提供了新型技术平台,对临床微血管血栓的治疗和预防具有潜在的应用价值。To study the formation and evolution of microvascular embolisms under different flow conditions and their effects on blood flow distribution,we designed and fabricated a microvascular chip based on Murray's law.Firstly,using microfluidic technology,a dynamic blood flow environment simulating physiological conditions was established.Then laser speckle contrast imaging was employed to obtain real-time blood flow field distributions.Finally,by analyzing the dynamic behavior of thrombus emboli and their impact on blood flow distribution,combined with hemodynamic simulation results,the role of hemodynamic factors on embolization was revealed.The results showed that microvascular bifurcation vertices,where blood flow velocity changed and wall shear stress decreased,tended to facilitate thrombus accumulation and growth.When emboli reached a critical size,they might detach and cause mechanical occlusion in downstream branches.Although collateral circulation could maintain the stability of outlet flow velocity in the case of incomplete obstruction,increased flow velocity in collateral vessels could still lead to vascular wall damage and impair local tissue blood exchange efficiency.This research provides a novel technical platform for investigating the mechanisms of microvascular embolism,and holds potential clinical value for the treatment and prevention of microvascular thrombosis.
