八子补肾胶囊对果蝇的抗衰老作用及机制

摘  要:目的探讨八子补肾胶囊对雄性果蝇的抗衰老作用及其机制。方法将雄性黑腹果蝇随机分为对照组、300 mg·L^(-1)八子补肾胶囊干预组、800 mg·L^(-1)八子补肾胶囊干预组、1200 mg·L^(-1)八子补肾胶囊干预组,每组100只。对照组果蝇在基础培养基中培养,300、800、1200 mg·L^(-1)八子补肾胶囊干预组果蝇分别在对应浓度的含药培养基中培养。记录果蝇生存情况并计算生存率、最长寿命、延寿率、平均寿命;在培养第15天和第30天评估果蝇的运动能力;监测果蝇繁殖能力并计算羽化率;采用比色法检测果蝇体内超氧化物歧化酶(SOD)、过氧化氢酶(CAT)的活性以及丙二醛(MDA)含量;评估冷应激条件下果蝇的抗应激损伤能力,并计算苏醒率。结果与对照组相比,800、1200 mg·L^(-1)八子补肾胶囊干预组果蝇的生存率显著提高(P<0.05);但不同浓度干预组间果蝇生存率比较差异无统计学意义(P>0.05);对照组及300、800、1200 mg·L^(-1)八子补肾胶囊干预组果蝇的最长寿命分别为68、73、77、79 d,300、800、1200 mg·L^(-1)八子补肾胶囊干预组果蝇的延寿率分别为9.70%、16.07%及11.24%;4组果蝇的平均寿命比较差异无统计学意义(P>0.05)。在培养第15天和第30天时,800 mg·L^(-1)八子补肾胶囊干预组果蝇高攀爬区域占比均显著高于对照组(P<0.05);且在第15天时,1200 mg·L^(-1)八子补肾胶囊干预组果蝇高攀爬区域占比显著高于300 mg·L^(-1)八子补肾胶囊干预组(P<0.05)。300、800、1200 mg·L^(-1)八子补肾胶囊干预组果蝇羽化率均显著高于对照组(P<0.05);800 mg·L^(-1)八子补肾胶囊干预组羽化率显著低于300 mg·L^(-1)八子补肾胶囊干预组(P<0.05)。与对照组相比,300、800、1200 mg·L^(-1)八子补肾胶囊干预组果蝇CAT活性显著增加(P<0.05),MDA含量显著降低(P<0.05),800 mg·L^(-1)八子补肾胶囊干预组果蝇中SOD活性显著增加(P<0.05)。在冷应激条件下,800、1200 mg·L^(Objective To investigate the anti-aging effect of Bazi Bushen capsules on male Drosophila melanogaster and its mechanisms.Methods Male Drosophila melanogaster were randomly divided into 4 groups(a control group,300,800,and 1200 mg·L^(-1)Bazi Bushen capsule groups,n=100).Drosophila melanogaster in the control group were cultured in a basic medium,while those in 300,800,and 1200 mg·L^(-1)Bazi Bushen capsule groups were cultured in medicated media containing the corresponding concentrations of the drug.The survival status of Drosophila melanogaster was recorded,and the survival rate,maximum lifespan,lifespan extension rate,and average lifespan were calculated.Locomotion ability was assessed on the 15 th and 30 th days after culture.Reproductive capacity was monitored and the eclosion rate was calculated.The activities of superoxide dismutase(SOD)and catalase(CAT)and the malondialdehyde(MDA)content in Drosophila melanogaster were measured by colorimetry.The anti-stress ability and the recovery rates of Drosophila melanogaster under cold stress were evaluated.Results Compared with that of the control group,the survival rate of 800 and 1200 mg·L^(-1)Bazi Bushen capsule intervention groups were significantly increased(P<0.05).However,there was no significant difference in survival rate among the three Bazi Bushen capsule groups(P>0.05).The maximum lifespan of Drosophila melanogaster in the control,300,800,and 1200 mg·L^(-1)Bazi Bushen capsule groups was 68,73,77,and 79 days,respectively.The lifespan extension rates of 300,800,and 1200 mg·L^(-1)Bazi Bushen capsule groups were 9.70%,16.07%,and 11.24%,respectively.No significant differences were observed in average lifespan among the four groups(P>0.05).The proportion of high-climbing area in the 800 mg·L^(-1)Bazi Bushen capsule group was significantly higher than that in the control group on both the 15 th and 30 th days after culture(P<0.05).The proportion of high-climbing area in the 1200 mg·L^(-1)Bazi Bushen capsule group was significantly higher than that in th
