UPLC-HRMS联合网络药理学探究隔山消醇提物治疗STC的物质基础及作用机制

摘  要:目的探讨土家族药物隔山消醇提取物治疗慢性传输型便秘(STC)的物质基础及可能的作用机制。方法采用超高效液相色谱-高分辨质谱法(UPLC-HRMS)技术分析隔山消60%乙醇提取物中的化学成分;从UPLC-HRMS检测获得的化学成分中,选取其中含量TOP20的化学成分结合PubChem、SITCH、TTD等数据库检索、筛选隔山消醇提取物治疗STC的作用靶点,BioGPS数据库对靶点对应的组织分布进行分析,利用微生信在线网站进行GO功能、KEGG通路的富集分析,筛选其治疗STC可能的主要活性成分及作用机制。结果UPLC-HRMS共检测出隔山消醇相对含量TOP20的化学成分,其相对含量约为总含量的60%,预测出186个隔山消醇提取物治疗STC的共同作用靶点。GO富集分析发现隔山消醇提取物化学成分治疗STC涉及ERK1和ERK2级联的正向调节、MAPK级联的正向调控、细胞增殖的正向调控等生物过程。KEGG富集通路共159条,包括AGE-RAGE信号通路、C型凝集素受体信号通路、PI3K-Akt信号通路、Ras信号通路、Rap1信号通路等。结论隔山消醇提取物中发挥治疗STC的可能化学成分为Meldenin、28-没食子酰葡萄糖基儿茶素-3-阿拉伯糖苷、9,10,11-三羟基十八碳-12-烯酸等,其作用机制可能与调节AGE-RAGE信号通路、C型凝集素受体信号通路、PI3K-Akt信号通路有关。Objective To investigate the material basis and potential mechanism of Cynanchum wilfordiiGeshanxi alcohol extract,a traditional Tujia medicine,in the treatment of slow transit constipation(STC).Methods Ultra performance liquid chromatography-high resolution mass spectrometry(UPLC-HRMS)was used to analyze the chemical components in the 60%ethanol extract of Cynanchum wilfordiiGeshanxi.From the chemical components obtained by UPLC-HRMS,the TOP20 chemical components were selected,and the PubChem,SITCH,TTD and other databases were used to search and screen the targets for the treatment of STC with the extract of Cynanchum wilfordiiGeshanxi.The BioGPS database was used to analyze the tissue distribution corresponding to the targets.Microbial informatics online platform was utilized for enrichment analysis of GO functions and KEGG pathways to screen the potential main active components and mechanisms of Cynanchum wilfordiiGeshanxi alcohol extract in the treatment of STC.Results The top 20 chemical components were detected in the Cynanchum wilfordiiGeshanxi alcohol extract by UPLC-HRMS,and the top 20 components accounted for approximately 60%of the total.A total of 186 common targets of Cynanchum wilfordiiGeshanxi alcohol extract in the treatment of STC were predicted.GO enrichment analysis revealed that the chemical components of Cynanchum wilfordiiGeshanxi alcohol extract involved in the treatment of STC are related to biological processes such as positive regulation of ERK1 and ERK2 cascade,positive regulation of MAPK cascade,and positive regulation of cell proliferation.A total of 159 KEGG enrichment pathways were identified,including AGE-RAGE signaling pathway,C-type lectin receptor signaling pathway,PI3K-Akt signaling pathway,Ras signaling pathway and Rap1 signaling pathway.Conclusion The potential chemical components in Cynanchum wilfordiiGeshanxi alcohol extract that may contribute to the treatment of chronic transit constipation(STC)include Meldenin,28-Galloylglucosylpomolate 3-arabinoside,and 9,10,11-Trihydro
