摘 要:【目的】系统鉴定和分析花生(Arachis hypogaea)类甜蛋白(TLP)基因家族,为花生TLP基因的抗旱功能研究及抗旱新品种培育提供参考。【方法】基于花生现有的基因组数据,利用HMM Search、blastp对花生TLP基因家族成员进行鉴定,利用生物信息学软件及程序分析花生TLP蛋白的理化性质、染色体定位、系统进化关系、保守基序及结构域等。将耐旱花生品种(L422)和干旱敏感品种(L677)在播种后45 d进行控水处理,以正常浇水为对照,分别在控水后5、7、9 d采集对照组、控水处理组花生植株组织进行转录组测序,分析花生TLP基因的干旱胁迫响应情况及组织表达模式。【结果】在花生20条染色体上鉴定到60个TLP基因,分别是AhTLP1~AhTLP60;AhTLP蛋白的氨基酸序列长度为225~543 aa,分子量为24.02~58.14 kD,等电点为4.14~9.37。依据系统进化关系,AhTLP蛋白分为7个类群,类群Ⅳ中成员数目最多,为14个,类群Ⅲ、Ⅵ中成员数目最少,为5个。AhTLP基因家族成员均包含thaumatin结构域及保守基序motif1、motif2、motif7、motif8;AhTLP5、AhTLP6、AhTLP8、AhTLP32、AhTLP33、AhTLP35、AhTLP47基因的表达能够被干旱诱导。【结论】花生基因组中鉴定到60个AhTLP基因,所有成员均含有thaumatin结构域,AhTLP5、AhTLP6、AhTLP8、AhTLP32、AhTLP33、AhTLP35、AhTLP47基因的表达能够响应干旱胁迫,可作为AhTLP基因功能研究的候选基因。【Objective】The study aimed at systematically identifying and analyzing TLP gene family from the cultivated peanut genome,and providing reference for functional analysis of peanut TLP genes in drought tolerance and cultivation of new drought-resistant varieties.【Method】Based on the existing genome data of peanut,HMM Search and blastp were used to identify the TLP family members of peanut.Bioinformatics software and programs were used to analyze the physicochemical properties,chromosome localization,phylogenetic relationships,conserved motifs and domains of the peanut TLP.The drought-tolerant variety L422 and the drought-sensitive variety L677 were subjected to water control treatment at 45 days after planting,with normal watering as the control.Plant tissues from the control group and treatment group were collected at 5,7 and 9 days after water control for transcriptome sequencing,and drought stress response and tissue expression patterns of the peanut TLP genes were analyzed.【Result】60 TLP genes were identified on 20 chromosomes of peanut,including AhTLP1-AhTLP60;The range of amino acid sequence length,molecular weight and isoelectric point of AhTLP was 225-543 aa,24.02-58.14 kD,4.14-9.37,respectively;Phylogenetic analysis showed that AhTLP was divided into 7 groups.The maximum number of members in Group IV was 14,while the minimum number in Groups III and VI was 5;All of the AhTLP members contained the thaumatin domain and conserved motifs 1,motifs 2,motifs 7,motifs 8;The expression levels of AhTLP5,AhTLP6,AhTLP8,AhTLP32,AhTLP33,AhTLP35 and AhTLP47 could be induced by drought stress.【Conclusion】There are 60 AhTLP genes in the peanut genome,all of which containes the thaumatin domain.The expression levels of AhTLP5,AhTLP6,AhTLP8,AhTLP32,AhTLP33,AhTLP35 and AhTLP47 can respond to drought stress,and therefore they can be the main target genes of functional analysis.
