摘  要:【目的】为进一步探究ARF基因在圆齿野鸦椿生长与发育过程中的作用机制提供科学依据。【方法】通过圆齿野鸦椿全基因组,利用生物信息学,鉴定其ARF基因家族成员,分析EkARF基因的理化性质、染色体定位、进化关系、基因结构、所包含的结构域以及顺式作用元件。探究EkARF基因在不同组织中的表达模式并进行功能预测。【结果】共鉴定出29个EkARF基因(EkARF1~EkARF29),分为4个亚族;除LG02染色体外,其余染色体上均有分布。29个ARF基因的氨基酸数量介于432~1110个,相对分子质量在48159.66~122724.62 u,均为亲水性蛋白,其亚细胞定位预测均在细胞核内。29个EkARF蛋白均含有DBD、Auxin_resp结构域,其中21个含有CTD结构域,其基因启动子顺式作用元件与植物光响应、激素的调节、防御机制和生长发育密切相关。EkARF基因在不同组织中表达存在差异,EkARF10在种子中高表达,可能参与种子发育调控;EkARF5随着果实的成熟表达量逐渐降低,可能在果实发育过程中起重要调控作用;EkARF16、EkARF20随着果实发育表达量上调,尤其是EkARF20在果实脱落期呈高表达,与果实成熟变红后仍挂果超130 d不脱落的现象一致,可能是果实发育的关键调控基因。【结论】本研究揭示了EkARF基因在圆齿野鸦椿不同组织部位中的表达模式,为阐明EkARF基因在圆齿野鸦椿生长发育过程中的功能提供理论基础。【Objective】The paper aimed to provide a reference for further investigation of the ARF gene in the growth and development of Euscaphis konishii Hayata.【Method】In the study,we had identified the members of the ARF gene family in E.konishii based on its genome.We conducted bioinformatics analysis and functional prediction of the EkARF gene,including the basic properties,chromosomal localization,evolutionary tree,gene structure,structural domains,cis-acting elements,and expression patterns in different tissues of EkARF gene family members using bioinformatics tools.【Result】A total of 29 EkARF genes(EkARF1-EkARF29)were identified and divided into four subfamilies;They were distributed on all chromosomes except chromosome LG02,the amino acid counts ranged from 432 to 1110,the relative molecular masses from 48159.66 to 122724.62 u.All of them were hydrophilic proteins.Subcellular localization was predicted to be in the nucleus.All 29 proteins contained DBD and Auxin_resp structural domains,and 21 of them contained CTD structural domains.The cis-acting elements were assoicated with light response,phytohormones,defense and stress,and plant growth and development.The 29 EkARF genes were differentially expressed in various tissues.EkARF10 was highly expressed in seeds and might be involved in the regulation of seed development;EkARF5 expression decreased gradually with fruit maturation and might play an important role in fruit development;EkARF16 and EkARF20 were up-regulated with fruit development,especially EkARF20 was highly expressed during fruit abscission,which was consistent with the strange phenomenon that the fruits were still hanging up for more than 130 days after maturation and turning red.The high expression of EkARF20 during fruit abscission was consistent with the strange phenomenon that the fruits remained hanging for up to 130 days after maturing and turning red,which might be the key regulatory genes for fruit development.【Conclusion】The study revealed the expression pattern of EkARF gene
