摘  要:【目的】旨在探索三叶青不同种植模式对土壤微生物群落结构的影响,为优化林下三叶青生态种植生产提供理论支撑。【方法】通过设置林下袋栽和地栽2种种植模式,一年的生长期后,测定三叶青根际土壤理化性质,结合Illumina MiSeq测序技术分析不同模式下根际土壤微生物物种组成、多样性和群落构建的差异。【结果】三叶青袋栽和地栽2种种植模式根际土壤的理化性质和群落结构具有明显差异。土壤总磷、总钾和有效磷差异极显著。对于细菌和真菌样品中的OTU数目,均是袋栽模式高于地栽模式。在门水平上,地栽模式下,土壤优势细菌群落是变形菌门(Proteobacteria,29%)、酸杆菌门(Acidobacteriota,26%);袋栽模式下,土壤优势细菌群落是酸杆菌门(Acidobacteriota,24%)、变形菌门(Proteobacteria,22%)和绿弯菌门(Chloroflex,15%);地栽模式下,土壤优势真菌群落是担子菌门(Basidiomycota,49%)、子囊菌门(Ascomycota,16%);袋栽模式下是子囊菌门(Ascomycota,46%)、担子菌门(Basidiomycota,26%)。【结论】三叶青不同种植模式会对根际土壤理化性质和土壤微生物群落造成不同影响,土壤pH等指标形成的微环境空间的差异可能是造成2种模式土壤群落组成差异的主要因素。【Objective】The study aimed to explore the effects of different planting patterns of Tetrastigma hemsleyanum on soil microbial community structure,providing theoretical support for optimizing the ecological cultivation of T.hemsleyanum under forest.【Method】Two planting patterns were set up:Bag planting and ground planting.After one year of growth,the physico-chemical properties of the T.hemsleyanum rhizosphere soil were measured.The differences in the species composition,diversity and community structure of soil microorganisms between the roots of different modes were analyzed with Illumina MiSeq sequencing technology.【Result】Physico-chemical properties and community structure of the rhizosphere soils between the two planting patterns of T.hemsleyanum under forest,were significantly different.The differences in total soil phosphorus,total potassium and effective phosphorus were highly significant.For the number of OTUs in the bacterial and fungal samples,it was higher in the bag planting pattern than in the ground planting pattern,and the dominant bacterial phyla of soil in the ground planting pattern were Proteobacteria(29%)and Acidobacteriota(26%),while the dominant bacterial phyla of soil in the bag planting pattern were Acidobacteriota(24%),Proteobacteria(22%)and Chloroflexi(15%);For fungal communities,under the ground cultivation pattern,the dominant groups were Basidiomycota(49%)and Ascomycota(16%).Under the bag cultivation pattern,the dominant groups were Ascomycota(46%)and Basidiomycota(26%).【Conclusion】Different planting modes of T.hemsleyanum have distinct effects on the physicochemical properties of the rhizosphere soil and the soil microbial community.The differences in microenvironmental spaces formed by factors such as soil pH may be the primary reason for the variations in soil community composition between the two planting patterns.
