摘 要:【目的】了解苍山潜穴虻(Vermiophis cangshanensis Li, Zhao&Wang, 2024)的线粒体基因组特征,进一步确立苍山潜穴虻的分类归属,探究苍山潜穴虻与穴虻科其他昆虫的系统发育关系。【方法】利用Illumina Novaseq 6000二代测序技术测定苍山潜穴虻线粒体全部基因组,对其进行注释和分析;选取线粒体细胞色素氧化酶I基因(cox1)序列,应用贝叶斯法(Bayesian inference, BI)构建系统发育树。【结果】苍山潜穴虻线粒体基因组(GenBank登录号:PP454204)全长为17 428 bp,其中非编码区(D-Loop区)和编码区共编码37个基因,编码区共包括蛋白质编码基因13个,rRNA基因2个和tRNA基因22个。平均测序深度为3076.48 X,GC含量为20.0%,碱基含量分别为A 39.0%、C 12.0%、G 8.0%、T 41.0%。除trnS1外,其余21个tRNA的二级结构均为典型的三叶草结构。【结论】本研究结果支持穴虻科和潜穴虻属的单系性,苍山潜穴虻确应置于潜穴虻属。【Objective】The present paper aimed to elucidate the mitochondrial genome characteristics of Vermiophis cangshanensis Li, Zhao & Wang, 2024,and further establish the classification and attribution of Vermiophis,and examine the phylogenetic relationship of the V.cangshanensis and other members of Vermileonidae.【Method】The mitochondrial genome of this species was sequenced using Illumina Novaseq 6000 next-generation sequencing technology, annotated, and analyzed.The phylogenetic tree was constructed based on the mitochondrial cytochrome c oxidase I(cox1) sequences under Bayesian Inference(BI).【Result】The mitochondrial genome of V.cangshanensis(GenBank accession number: PP454204) measure 17 428 bp in length, including one non-coding(D-loop) region and 37 genes, and the coding region contained 13 PCGs, 22 tRNA genes, 2 rRNA genes.The average sequencing depth was 3076.48 X.The GC content was 20.0%,and the individual base contents were as follows: A(39.0%),C(12.0%),G(8.0%) and T(41.0%).Except for trnS1,the secondary structures of the other 21 tRNAs were a typical clover-leaf structure.【Conclusion】According to the phylogenetic analysis, it can be drawn that the monophyly is supported for both Vermelionidae and Vermiophis,and the species V.cangshanensis is supported to be a member of the genus Vermiophis.
