电针干预对慢性神经压迫损伤模型小鼠痛行为的影响

摘　　要：目的:观察电针疗法对慢性神经压迫损伤(CCI)模型小鼠痛行为的影响。方法:随机把40只C57BL/6小鼠分成对照组、模型组、电针组和假电针组,每组各10只。采用含铬羊肠线结扎坐骨神经的方法,建立CCI动物模型。电针组和假电针组分别在造模后进行电针和假电针的干预7天,其余组不予干预。观察造模后各组小鼠行为学的变化情况。结果:与对照组比较,术后7天至28天造模组小鼠痛阈均明显降低(P﹤0.05,P﹤0.01),术后14天焦虑情绪改变(P﹤0.05,P﹤0.01),术后21天抑郁情绪改变(P﹤0.05,P﹤0.01),且均持续到术后28天。通过电针和假电针的干预,与模型组比较,小鼠痛阈得到提升,焦虑、抑郁情绪得到缓解(P﹤0.05,P﹤0.01);且电针组疗效更佳(P﹤0.05,P﹤0.01)。结论:电针干预对于小鼠慢性疼痛伴发焦虑抑郁情绪具有良性作用。Objective:To observe the effect of electroacupuncture therapy on painful behavior of model mice with chronic constriction injury(CCI).Methods:40 C57BL/6 mice were randomly divided into 4 groups:control group,model group,electroacupuncture group and sham electroacupuncture group,10 mice in each group.The animal model of CCI was established by ligating sciatic nerve with chromium-containing catgut.After successful modeling,electroacupuncture group and sham electroacupuncture group were intervened respectively by electroacupuncture and sham electroacupuncture for 7 days,while other groups were not intervened.The behavioral changes of mice were observed in each group after modeling.Results:Compared with the control group,the pain threshold of mice in model group was significantly reduced from 7 days to 28 days after operation(P﹤0.05,P﹤0.01);the anxious mood was changed on the 14th day after operation(P﹤0.05,P﹤0.01),the depressive mood was changed on the 21th day after operation(P﹤0.05,P﹤0.01),and which all lasted until the 28th days after operation.Through the interventions by electroacupuncture and sham electroacupuncture,compared with the model group,the pain threshold of mice was elevated,and anxious and depressive moods were alleviated(P﹤0.05,P﹤0.01),and the clinical efficacy of electroacupuncture group was better(P﹤0.05,P﹤0.01).Conclusion:Electroacupuncture intervention has a benign effect on mice's chronic pain accompanied by anxious and depressive moods.
