消镇康复方的提取工艺优化及其抗结肠癌活性的考察

摘　　要：目的优化消镇康复方的提取工艺,考察优化提取物的小鼠血清对结肠癌细胞的抑制能力。方法在单因素试验的基础上,选取乙醇浓度、提取时间、提取次数、料液比四个因素进行L9(3^(4))正交试验,以提取液总皂苷、总生物碱含量为综合评价指标,优化消镇康复方的提取工艺,获得的优化提取液小鼠灌胃后,收集的血清对结肠癌细胞HCT-116开展活性实验,比较抑制率。结果消镇康复方的最佳提取工艺为:乙醇体积浓度70%、提取次数2次、提取时间2 h,料液比1∶8;正交优化提取物对HCT-116细胞有明显抑制作用,且效果优于水煎煮提取物。结论消镇康复方提取工艺的优化方法准确可行,优化提取物抑制结肠癌细胞的活性强于水煎煮提取物,为消镇康胶囊的新药研发奠定基础,为抗结肠癌中药复方及其制剂的研究提供有益的思路。Objective To optimize the extraction process of Xiaozhenkang formula and to investigate the inhibitory ability of optimized mouse serum on colon cancer cells.Methods According to the single factor test,four factors were selected to carry out L9(34)orthogonal test,namely:ethanol concentration,extraction time,number of times of extraction,and material-liquid ratio.Then the contents of total saponins and total alkaloids in the extract were used as comprehensive evaluation indexes to optimize the extraction process of the elimination and rehabilitation formula.After the extract of the optimized process was used in the gavage of mice,the collected serum was used to carry out activity experiments on colon cancer cells HCT-116,and the inhibition rates of the optimized group and the water extraction group were compared.Results The optimal extraction process of the Xiaozhenkang formula was as follows:extracted with 8 times the amount of 70%ethanol for 2 times,2 h each time;orthogonally optimized extracts had a significant inhibitory effect on HCT-116 cells,and the effect was better than that of water decoction extract.Conclusion The optimization method for the extraction process of Xiaozhenkang formula is accurate and feasible.The activity of the optimized extract in inhibiting colon cancer cells is stronger than that of the water decoction extract,laying the foundation for the research of new drugs of Xiaozhenkang capsules and providing beneficial ideas for the research of traditional Chinese medicine prescriptions against colon cancer.
