摘  要:采用RT-PCR和RACE技术从爪哇根结线虫中克隆1个扩展蛋白基因的cDNA序列命名为Mj-expb1,该序列全长587 bp,ORF长度393 bp,编码蛋白长度130 aa。利用生物信息学软件对其基因结构、编码蛋白的系统进化进行分析。结果显示,该蛋白含有1个DPBB结构域,为带负电荷疏水性稳定蛋白,以丝氨酸磷酸化修饰调控为主,无跨膜结构;定位于细胞外,N端含有信号肽,为细胞外分泌蛋白,二级结构以无规则卷曲为主,三级结构为单体蛋白。此外,氨基酸序列比对中保守结构域DPBB在线虫中的同源性较高,系统发育树显示扩展蛋白在线虫的属间差异大,种间差异较小。RT-PCR and RACE techniques were used to clone a cDNA sequence of an expansin gene from Meloidogyne javanica,named Mj-expb1The full length of the cDNA sequence is 587 bp,with an ORF length of 393 bp,encoded protein length of 130 aaBioinformatics software was utilized to analyze the gene structure,encoded protein,and phylogenetic evolutionThe results show that the protein contains a DPBB domain,which is a negatively charged hydrophobic stable protein,is mainly regulated by serine phosphorylation modification without transmembrane structure,is located extracellularly,and the N-terminus contains a signal peptide,indicating it is an extracellular secretory proteinThe secondary structure is mainly composed of random coils,and the tertiary structure is a monomeric proteinIn the amino acid sequence alignment,the DPBB domain shows high homology in nematodesThe phylogenetic tree indicates that expansins have large differences among genera but small differences among species in nematodes.
