摘 要:帝王大丽花(Dahlia imperialis)是新近引入国内的珍稀花卉,常通过分切块根繁殖或待块根发芽后采芽扦插以获得种苗。但帝王大丽花块根萌芽率低,且块根无性繁殖多代后常因病原累积导致繁殖系数和种苗质量均显著下降;同时,帝王大丽花扦插繁殖中插穗褐变严重导致成活率低。能否通过组培诱导大丽花微型块根作为繁殖材料尚未见报导。本文通过正交试验,探索多因素影响下组培苗块根的诱导和发生。结果表明:1/2MS+0.5 mg·L^(-1)萘乙酸(NAA)+0.5 mg·L^(-1)吲哚丁酸(IBA)+光暗时间比4 h/20 h培养条件可诱导块根形成,根直径可达0.67 cm,接种后10 d时生根率便可达到100%。合适浓度的多效唑(PP_(333))处理能增加块根数量,抑制了块根增粗及组培苗增高。研究结果能为生产大丽花高质量的繁殖材料提供参考。Dahlia imperialis is a rare tuberous ffower recently introduced into China,and is often propagated by dividing the tuberous roots or taking bud cuttings after the tuberous roots sprouting in order to obtain seedlings.However,the adventitious buds rate of the tuberous roots of D.imperialis is low,and the reproduction coefffcient and seedling quality often decrease signiffcantly due to the accumulation of pathogens after several generations of asexual propagation of the tuberous roots.The survival rate of cuttings of D.imperialis was low due to serious browning.The possibility of inducing dahlia micro-tubers as propagation material through tissue culture has not been reported.This article explored the induction of root tubers in tissue culture seedlings of D.imperialis under multiple factors through orthogonal experiments.The results showed that under the culture conditions of 1/2MS+0.5 mg·L^(−1) NAA+0.5 mg·L^(−1) IBA+light-dark period ratio 4 h/20 h,micro-tubers can be induced with a diameter of 0.67 cm,achieving 100%rooting within just 10 d. The treatment of suitable concentration of paclobutrazol (PP_(333)) increased the number of tuberous roots and inhibited the thickening of roots and plantlets height. The results of the study can provide a referencefor the production of high-quality propagation materials for dahlias.
