茭白ZlPDS基因的克隆及其在肉质茎品质形成与采后呈色转变过程中的表达分析

摘  要:茭白肉质茎是由菰黑粉菌(Ustilago esculenta)侵染诱导菰(Zizania latifolia)植株茎部膨大发育形成的,但其表皮货架期易变黄,严重影响茭白品质。本研究采用光照处理茭白肉质茎,其类胡萝卜素各组分含量均显著上升, qPCR分析发现6个类胡萝卜素生物合成途径相关基因表达均显著升高。克隆获得八氢番茄红素脱氢酶基因(ZlPDS)全长序列,进化树分析发现其与粳稻PDS同源性最高,并受菰黑粉菌侵染诱导,在菰黑粉菌侵染15 d后的野茭植株茎部表达显著降低;茭白植株茎部起始膨大发育后, ZlPDS及其下游β-胡萝卜素羟化酶(ZlBCH)与类胡萝卜素ε-羟化酶(ZlECH)基因表达均显著降低,并在茎部膨大前后表达变化趋势一致,可能参与茭白肉质茎白色品质建成及维持调节。茭白植株起始于ZlPDS表达下调的类胡萝卜素生物合成可能是菰黑粉菌侵染诱导茭白肉质茎白色品质形成与维持的重要调节途径,本研究为菰黑粉菌侵染诱导茭白肉质茎品质形成的调节机制提供了新的研究思路。The ffeshy stem was formed by the infection of Ustilago esculenta,which induced the swelling development of the stem in Zizania latifolia.However,its epidermal was prone to yellowing in shelf life,seriously affecting the quality of normal Z.latifolia.After light exposure,the various components of carotenoids were signiffcantly increased in the swollen stems of normal Z.latifolia.Meanwhile,qPCR analysis found that the expression of 6 genes related to carotenoid biosynthesis pathways were signiffcantly increased.The full-length sequence of phytoene desaturase gene(ZlPDS)was cloned and found to have the highest homology with PDS in japonica rice by phylogenetic tree analysis.The expression in the stem of wild Z. latifolia was signiffcantly decreased after 15 days infection of U. esculenta. During the initial swelling and development of the stem in normal Z. latifolia, the expression of ZlPDS and its downstream genes of β-carotene hydroxylase (ZlBCH) and carotenoid ε-hydroxylase (ZlECH) were signiffcantly reduced. The expression trend was consistent before and after the swelling of stem, which may be involved in the regulationand maintenance of the white quality of ffeshy stem in normal Z. latifolia. The biosynthesis of carotenoidsstarting from the downregulation of ZlPDS would be an important regulatory way for the formation and maintenance of white quality in normal Z. latifolia induced by infection of U. esculenta. This reaserach provided a new research approach for the regulatory mechanism of the quality formation of the fleshy stem of Z. latifolia induced by the infection of U. esculenta.
