摘  要:目的探讨维生素D对白癜风小鼠免疫功能的影响以及对TLR4/MyD88/NF-κB信号通路的调节作用。方法将90只C57BL/6N小鼠随机数字表法分为对照组、模型组、TAK-242组(TLR4抑制剂,3 mg/kg)、维生素D低剂量组(50 ng/mL)、维生素D高剂量组(100 ng/mL),每组18只,除对照组外,其余各组小鼠通过涂抹莫诺苯宗乳膏构建小鼠白癜风模型。评估各组小鼠毛发脱色情况;测定脾和胸腺指数;ELISA检测小鼠血清中IL-1β、IL-17、IFN-γ的水平;流式细胞术检测小鼠外周血淋巴T淋巴细胞亚群水平;Western blot检测TLR4/MyD88/NF-κB信号通路相关蛋白表达。结果与对照组相比,模型组小鼠脾和胸腺指数、CD4^(+)及CD4^(+)/CD8^(+)水平降低(P<0.05),炎症因子IL-1β、IL-17、IFN-γ、CD8^(+)水平及TLR4、MyD88、NF-κB p65磷酸化蛋白表达升高(P<0.05);与模型组相比,TAK-242组和维生素D低、高剂量组上述指标均相反表达(P<0.05)。结论维生素D可以改善白癜风小鼠的免疫功能,其作用机制可能与抑制TLR4/MyD88/NF-κB信号通路有关。Objective To investigate the effects of vitamin D on immune function and the regulation of TLR4/MyD88/NF-κB signaling pathway in vitiligo mice.Methods 90 C57BL/6N mice were randomly divided into control group,model group,TAK-242 group(TLR4 inhibitor,3 mg/kg),vitamin D low-dose group(50 ng/mL)and vitamin D high-dose group(100 ng/mL)with 18 mice in each group.In addition to the control group,the vitiligo model was constructed by applying monobenzone cream in other groups of mice.The hair decolorization of each group was evaluated.Spleen and thymus index were determined.Serum levels of IL-1β,IL-17 and IFN-γwere detected by ELISA.T lymphocyte subsets in peripheral blood of mice were detected by flow cytometry.Western blot were used to detect TLR4/MyD88/NF-κB signaling pathway related protein expression.Results Compared with the control group,the spleen and thymus indexes,CD4^(+)and CD4^(+)/CD8^(+)levels were decreased in model group(P<0.05),the levels of inflammatory cytokines IL-1β,IL-17,IFN-γ,CD8^(+)and the expression of TLR4,MyD88,NF-κB p65 phosphorylated protein were increased(P<0.05).Compared with model group,the above indexes were inversely expressed in TAK-242 group and low and high dose vitamin D groups(P<0.05).Conclusion Vitamin D can improve the immune function of vitiligo mice,and its mechanism may be related to inhibition of TLR4/MyD88/NF-κB signaling pathway.
