摘 要:目的基于磷酸盐(PO_(4)^(3-))调控的腺苷激酶(ADK)活性,探究黑磷纳米片(BPNSs)的选择性降解对ADK活性的调控。方法利用中性和酸性两种溶液分别模拟正常生理环境、肿瘤微环境,通过吸收强度及PO_(4)^(3-)含量测定探究BPNSs在两种溶液中的降解特性;并通过酶促反应动力学探究BPNSs在两种溶液中对ADK催化腺苷磷酸化反应的调控。结果BPNSs在模拟的肿瘤微环境中更易降解,产生的PO_(4)^(3-)明显高于模拟的正常生理环境。ADK在两种溶液中均能催化腺苷磷酸化,然而其活性受高浓度腺苷抑制。受益于在模拟的肿瘤微环境中降解产生高含量的PO_(4)^(3-),BPNSs恢复了ADK活性并促进了ADK催化的腺苷转化。结论基于黑磷纳米材料降解特性和酶活性调控机制,BPNSs有望成为新型腺苷代谢调控药物和腺苷介导的免疫抑制调节剂。Objective To investigate the selective degradation of black phosphorus nanosheets(BPNSs)on the regulation of adenosine kinase(ADK)activity regulated by phosphate(PO_(4)^(3-)).Methods Neutral and acidic solutions were used to simulate normal physiological environment and tumor microenvironment,respectively.The degradation of BPNSs in both solutions were explored by measuring absorption intensity and PO_(4)^(3-).The regulation of BPNSs on ADK catalyzed adenosine phosphorylation in both solutions was investigated through the kinetic of enzyme-catalyzed reaction.Results In the simulated tumor microenvironment,BPNSs were degraded more easily and produced higher concentrations of PO_(4)^(3-).ADK could catalyze adenosine phosphorylation in both solutions,but its activity was affected by high concentration of adenosine.Benefiting from the degradation to produce high level of PO_(4)^(3-),BPNSs restored ADK activity and promoted ADK-catalyzed adenosine transformation in the simulated tumor microenvironment.Conclusion Based on the degradation of black phosphorus and the regulation mechanism of enzyme activity,BPNSs may become novel adenosine metabolism regulating drugs and adenosine-mediated immunosuppressive regulators.
