摘  要:目的建立华北鸦葱的质量控制方法。方法采用薄层色谱法定性华北鸦葱中的芦丁;以亚硝酸钠-硝酸铝-氢氧化钠为显色剂,采用紫外-可见分光光度法测定华北鸦葱中总黄酮的含量,检测波长为508 nm。结果薄层鉴别结果显示,供试品色谱中在与芦丁对照品色谱相应的位置上显相同颜色斑点。含量测定结果显示,芦丁在7.86~47.18 mg·L^(-1)范围内与吸光度呈良好的线性关系,回归方程为A=0.0132 C-0.0021(r=0.9999),其中C为芦丁对照品的质量浓度(mg·L^(-1)),A为吸光度,平均加样回收率为99.12%(RSD=1.53%)。结论本研究方法操作简便、准确高、重复性好,可作为华北鸦葱的质量控制方法。Objective To establish the quality control standard of Scorzonera albicaulis Bunge.Methods A thin-layer chromatography method was used to identify rutin in Scorzonera albicaulis Bunge.With nitrite-aluminum nitrate-sodium hydroxide as the chromogenic reagent,the content of the total flavonoids in Scorzonera albicaulis Bunge was detected by ultraviolet-visible spectrophotometry at 508 nm.Results The result of thin-layer chromatography identification showed that there was a spot of the same color in the chromatographic position corresponding to the chromatographic position of rutin control.The content determination result showed that there was a better linear relationship between concentration of rutin and absorbance in the range of 7.86-47.18 mg·L^(-1).The linear regression equation was A=0.0132 C-0.0021(r=0.9999),in which the C was the mass concentration of rutin reference(mg·L^(-1)),A was the absorbance.The average recovery of rutin was 99.12%(RSD=1.53%).Conclusion The research method is simple,accurate and repeatable,which can be used for the quality control standard of Scorzonera albicaulis Bunge.
