摘 要:目的探究太赫兹辐射对创面的作用效应及其潜在机制。方法自2023年12月至2024年5月,陆军军医大学第二附属医院(新桥医院)整形外科将16只6~8周龄C57BL/6J小鼠随机分为辐照组和对照组,8只/组,建立急性皮肤全层缺损创面模型。辐照组创面接受平均功率密度为110 mW/cm2的太赫兹暴露,暴露时间为10 min/孔,20 min/只,对照组创面暴露于同实验环境下的空气中,20 min/只。于辐照完成后即刻、第1天、第3天拍照记录创面愈合情况并通过Image J量化分析残留面积,采用HE染色对创面宽度、表皮平均厚度、肉芽组织厚度及面积进行分析,Masson染色和天狼星红染色分析创面胶原含量及类型,并对IL-10、IL-1β、CD31、CollagenⅢ、Ki-67等指标进行免疫组化染色分析。结果创面残留面积的半定量分析表明,辐照组小鼠创面愈合率在第1天和第3天均高于对照组,且两组第1天差异有统计学意义(P<0.01)。HE染色显示,第1天和第3天辐照组新生表皮长度和平均厚度均高于对照组,且在第3天时差异有统计学意义(P<0.0001)。Masson染色显示,辐照组新生胶原占比均高于对照组,且在创缘部位的占比差异有统计学意义(P<0.0001)。天狼星红染色显示,创缘部位辐照组新生Ⅲ型胶原/Ⅰ型胶原占比高于对照组,差异有统计学意义(P<0.05)。免疫组化结果提示,辐照组IL-1β阳性面积占比低于对照组,差异无统计学意义(P>0.05);辐照组IL-10、CollagenⅢ、CD31、Ki-67阳性面积占比均显著高于对照组,差异有统计学意义(P<0.0001,P<0.05,P<0.01,P<0.0001)。结论太赫兹辐射能够减轻炎症反应,促进创面新生血管形成,并促进小鼠皮肤创面胶原生成和再上皮化。Objective Toinvestigatetheeffectofterahertz radiation on acute wounds and its potential mechanism.Methods Six teen 6 to 8 weeks C57BL/6 J mice were randomly divided into the irradiated group and control group,8 mice in each group,to establish an acute whole skin defect trauma model.The wounds of the irradiated group received terahertz exposure with an average power density of 110 mW/cm2 for 10 min/hole(20 min/each),and the wounds of the control group were exposed to air in the same experimental environment for 20 min/each.Wound healing was recorded by taking photos immediately after exposure,on day 1 and day 3,and the residual area was quantified and analyzed by Image J.The width of the wound,average thickness of epidermis,thickness and area of granulation tissue were analyzed by HE staining,and the collagen content and type of the wound were analyzed by Masson staining and Sirius red staining.Im munohistochemical staining was performed to analyze IL-10,IL-1β,CD31,CollagenⅢ,and Ki-67 indexes.Results The semi-quanti tative analysis of the residual wound area showed that the wound healing rate of the irradiated group was higher than that of the control group on both day 1 and day 3,and the difference was significant on day 1(P<0.01).HE staining showed that the length and average thick ness of the new epidermis in the irradiated group were higher than those in the control group on days 1 and 3,and the difference was signif icant on day 3(P<0.0001).Masson staining showed that the percentage of new collagen in the irradiated group was higher than that in the control group,and the difference was significant at the edge of the wound(P<0.0001).Sirius red staining showed that the proportion of typeⅢcollagen/typeⅠcollagen was higher in the irradiated group than in the control group(P<0.05).Immunohistochemistry results suggested that the percentage of IL-1βpositive area was lower in the irradiated group than in the control group,but the difference was not significant(P>0.05).The percentage of IL-10,CollagenⅢ,CD31,
