补肾活血方调控Keap1/Nrf2/ARE通路减缓LPS诱导流产小鼠氧化应激的研究

摘  要:目的探讨补肾活血方通过调节Keap1/Nrf2/ARE通路减轻LPS诱导流产小鼠氧化应激的作用机制。方法将32只ICR孕鼠随机分为空白组、空白+补肾活血方组、模型组和模型+补肾活血方组。采用尾静脉注射LPS法诱导流产模型,同时进行补肾活血方灌胃干预。于妊娠第13天麻醉小鼠后解剖子宫,计算胚胎吸收率。采用HE染色观察蜕膜组织显微结构,ELISA检测外周血SOD和MDA含量,Western blot和RT-qPCR检测蜕膜组织Keap1、Nrf2、HO-1、NQO1和γ-GCS的表达。结果空白组与空白+补肾活血方组各项指标无明显差异。与空白组相比,模型组胚胎吸收率升高(P<0.05),蜕膜组织结构破坏,MDA、Keap1表达上升(P<0.05),SOD、Nrf2、HO-1和NQO1表达下降(P<0.05),γ-GCS表达无显著变化。与模型组相比,补肾活血方干预后胚胎吸收率降低(P<0.05),蜕膜组织结构改善,MDA、Keap1表达水平下降(P<0.05),SOD、Nrf2、HO-1和NQO1表达上升(P<0.05),γ-GCS表达无明显差异。结论补肾活血方可能通过激活Keap1/Nrf2/ARE信号通路减缓过度氧化应激反应,达到治疗RSA的作用。Objective To explore the effect of Bushen Huoxue Formula(BSHXF)in alleviating oxidative stress in LPS-induced abortion mice by regulating the Keap1/Nrf2/ARE signaling pathway.Methods 32 Institute of Cancer Research(ICR)pregnant mice were randomly divided into the blank group,the blank+BSHXF group,the model group and the model+BSHXF group.The tail vein injection of lipopolysaccharide(LPS)was used to induce the abortion model,and BSHXF was gavaged simultaneously.The uterus of mice was dissected after anesthesia on the 13th day of gestation and the embryo resorption rate was calculated.Hematoxylin-eosin(HE)staining was used to observe the microstructure of decidua tissues,enzyme-linked immunosorbent assay(ELISA)to detect the contents of SOD and MDA in peripheral blood,and Western blot(WB)and reverse transcription quantitative polymerase chain reaction(RT-qPCR)to test the expression levles of Keap1,Nrf2,HO-1,NQO1 andγ-GCS in the decidual tissues.Results There was no significant difference between the blank group and the blank+BSHXF group.Compared with the blank group,the embryo resorption rate in the model group was increased(P<0.05),the decidual tissue structure was damaged,the expressions of MDA and Keap1 was increased(P<0.05),the expressions of SOD,Nrf2,HO-1 was decreased(P<0.05),and the expression ofγ-GCS did not change significantly(P<0.05).Compared with the model group,the embryo resorption rate was decreased after the intervention of BSHXF(P<0.05),the decidual tissue structure was improved,the expression levels of MDA and Keap1 were decreased(P<0.05),the expression levels of SOD,Nrf2 and HO-1were increased(P<0.05),and the expression ofγ-GCS showed no significant difference.Conclusion BSHXF may alleviate excessive oxidative stress by activating the Keap1/Nrf2/ARE signaling pathway in treating recurrent spontaneous abortion(RSA).
