摘 要:目的探讨宫颈癌患者血清及组织中长链非编码RNA(lncRNA)CKMT2反义RNA 1(lncRNA CKMT2-AS1)、微小RNA(miR)-106a相对表达水平及其与人乳头瘤病毒(HPV)感染的关系。方法选取2021年4月至2023年4月该院收治的宫颈癌患者68例作为宫颈癌组,另取同时期该院收治的因患子宫肌瘤需行子宫全切术的患者68例作为对照组。采用实时荧光定量聚合酶链反应(qRT-PCR)检测2组患者血清lncRNA CKMT2-AS1、miR-106a相对表达水平,并检测宫颈癌患者癌组织和子宫肌瘤患者正常宫颈组织中lncRNA CKMT2-AS1、miR-106a相对表达水平;收集对照组、宫颈癌组患者的宫颈分泌物,检测2组患者HPV阳性情况;同时根据HPV载量变化情况将宫颈癌组HPV阳性患者分为HPV负荷量1~100组、HPV负荷量>100~1000组和HPV负荷量>1000组。采用Spearman相关分析血清及组织中lncRNA CKMT2-AS1、miR-106a相对表达水平与宫颈癌患者HPV感染负荷量的相关性。结果与对照组比较,宫颈癌组患者血清及组织中lncRNA CKMT2-AS1相对表达水平降低,miR-106a相对表达水平升高,差异均有统计学意义(P<0.05)。对照组和宫颈癌组患者HPV阳性率比较[1.47%(1/68)vs.95.59%(65/68)],差异有统计学意义(P<0.05)。宫颈癌患者中HPV负荷量1~100组28例,HPV负荷量>100~1000组21例,HPV负荷量>1000组16例。宫颈癌患者血清及组织中lncRNA CKMT2-AS1相对表达水平为HPV负荷量1~100组>HPV负荷量>100~1000组>HPV负荷量>1000组,且两两比较,差异均有统计学意义(P<0.05);宫颈癌患者血清及组织中miR-106a相对表达水平为HPV负荷量1~100组<HPV负荷量>100~1000组<HPV负荷量>1000组,且两两比较,差异均有统计学意义(P<0.05)。Spearman相关分析结果显示,宫颈癌患者血清及组织中lncRNA CKMT2-AS1相对表达水平与HPV负荷量呈负相关(P<0.001),miR-106a相对表达水平与HPV负荷量呈正相关(P<0.001)。结论宫颈癌患者血清及组织中lncRNA CKMT2-AS1相对表达水平相对较低,miR-Objective To investigate the relative expression levels of long non-coding RNA(lncRNA)CKMT2 antisense RNA 1(lncRNA CKMT2-AS1)and microRNA(miR)-106a in serum and tissues of patients with cervical cancer and their relationship with human papillomavirus(HPV)infection.Methods Sixty-eight patients with cervical cancer admitted to the hospital from April 2021 to April 2023 were selected as the cervical cancer group,and 68 patients with uterine fibroids who needed total hysterectomy in the hospital were selected as the control group.Real-time quantitative fluorescent PCR(qRT-PCR)was used to detect the relative expression levels of lncRNA CKMT2-AS1 and miR-106a in serum of the two groups,as well as the relative expression levels of lncRNA CKMT2-AS1 and miR-106a in cancer tissues of patients with cervical cancer and normal cervical tissues of patients with uterine fibroids.Cervical secretions of patients in the control group and the cervical cancer group were collected,and HPV positivity was detected in the two groups.At the same time,according to the changes of HPV load,HPV-positive patients in the cervical cancer group were subdivided into HPV load 1-100 group,HPV load>100-1000 group and HPV load>1000 group.Spearman correlation was used to analyze the correlation of the relative expression levels of lncRNA CKMT2-AS1 and miR-106a in serum and tissues with HPV infection load in patients with cervical cancer.Results Compared with the control group,the relative expression levels of lncRNA CKMT2-AS1 in serum and tissue of patients with cervical cancer group were decreased,and the relative expression levels of miR-106a were increased,with statistically significant differences(P<0.05).Comparison of HPV positivity rate between the control group[1.47%(1/68)]and the cervical cancer group[95.59%(65/68)]had statistically significant difference(P<0.05).Among the patients with cervical cancer,there were 28 cases in the HPV load 1-100 group,21 cases in the HPV load>100-1000 group and 16 cases in the HPV load>1000 group.The relative ex
