摘  要:目的利用UPLC-Q-TOF-MS/MS技术对利胆消痛方水煎液成分进行定性分析。方法采用Accucore C18色谱柱(100 mm×2.1 mm,2.6μm),柱温为40℃,流动相A为0.1%甲酸水,流动相B为乙腈,梯度洗脱。流速为0.4 mL·min^(-1),进样量为5μL。采用电喷雾离子源,在正、负离子模式下,精确分子质量和二级质谱裂解规律对检测到的化学成分进行鉴定,通过与对照品比对,结合文献报道,确定化学成分。结果在利胆消痛方中共鉴定出103个化学成分,包括48个黄酮类化合物,29个萜类化合物,8个有机酸类化合物,5个氨基酸类化合物,4个生物碱类化合物,4个香豆素类化合物和5个其他类化合物。结论本实验阐明利胆消痛方水煎液中的组成成分,为其药效物质基础、质量控制和作用机制研究提供了依据。Objective To qualitatively analyze the constituents of the aqueous decoction of Lidan Xiaotong formula with UPLC-Q-TOF-MS/MS technology,and to provide a basis for its quality control.Methods An Accucore C18 column(100 mm×2.1 mm,2.6μm)was used with 0.1%formic acid aqueous solution(A)-acetonitrile(B)as the mobile phase in a gradient elution mode.The column temperature was 40℃,and the flow rate was 0.4 mL·min^(-1)with the injection volume of 5μL.The electrospray ionization source was used to identify the detected chemical constituents by precise molecular mass and secondary mass spectrometry cleavage patterns in both positive and negative ion modes.The chemical constituents were determined by comparison with the controls and literature reports.Results Totally 103 chemical constituents,including 48 flavonoids,29 terpenoids,8 organic acids,5 amino acids,4 alkaloids,4 coumarins and 5 other compounds,were identified in Lidan Xiaotong formula.Conclusion This experiment elucidates the constituents in the aqueous decoction of Lidan Xiaotong formula,providing a reference for the study of its material basis,efficacy,quality control and mechanism of action.
