摘  要:目的建立UPLC-MS/MS法同时测定银黄胶囊中6个有机酸类(绿原酸、新绿原酸、隐绿原酸、3,5-O-二咖啡酰基奎宁酸、3,4-O-二咖啡酰基奎宁酸、4,5-O-二咖啡酰基奎宁酸)和4个黄酮类(黄芩苷、黄芩素、汉黄芩苷、汉黄芩素)成分的含量。方法采用Hypersil GOLD VANQUISH C_(18)柱(2.1 mm×100 mm,1.9μm),以乙腈-0.2%甲酸水溶液为流动相,梯度洗脱,柱温35℃,流速为0.3 mL·min^(-1);质谱采用电喷雾离子源,以多反应监测模式进行定量分析。结果10种成分在一定浓度范围内与峰面积线性关系良好(r>0.9990);平均加样回收率(n=3)均在97.35%~99.26%,RSD在0.36%~0.83%。6批银黄胶囊中上述10种成分含量分别为6.981~12.56、0.8929~2.084、1.407~3.454、0.5611~0.9415、0.8823~1.647、1.984~3.465、94.16~135.2、1.768~3.214、7.985~10.06、6.487~10.11 mg·g^(-1)。结论该方法快速准确,稳定灵敏,可用于银黄胶囊的质量控制。Objective To simultaneously determine 6 organic acids(chlorogenic acid,neochlorogenic acid,cryptochlorogenic acid,3,5-O-dicaffeoyl quinic acid,3,4-O-dicaffeoyl quinic acid and 4,5-O-dicaffeoyl quinic acid)and 4 flavonoids(baicalin,baicalein,wogonoside and wogonin)in Yinhuang capsules by UPLC-MS/MS.Methods The separation was performed on an Hypersil GOLD VANQUISH C_(18)(2.1 mm×100 mm,1.9μm).The mobile phase consisted of acetonitrile-0.2%formic acid by gradient elution at the column temperature of 35℃.The flow rate was 0.3 mL·min^(-1).Electrospray ionization source was used for the quantitative analysis in multiple reaction monitoring modes.Results Ten components showed good linearity with all correlation coefficients>0.9990.The average recoveries(n=3)ranged from 97.35%to 99.26%with RSD from 0.36%to 0.83%.The contents of the above 10 components in the 6 batches of Yinhuang capsules were within 6.981~12.56,0.8929~2.084,1.407~3.454,0.5611~0.9415,0.8823~1.647,1.984~3.465,94.16~135.2,1.768~3.214,7.985~10.06 and 6.487~10.11 mg·g^(-1),respectively.Conclusion The method is simple,accurate,reliable and sensitive for the quality control of Yinhuang capsules.
