摘 要:花生是我国重要的经济油料作物.为研究花生根系在干旱复水生长中的生理特征变化,研究对花生幼苗进行干旱处理,复水后观察花生根系生长情况,分析根系生长相关指标的变化,采用台盼蓝染色检测根尖细胞存活状态,并进一步分析AhmTERF1表达变化,探讨AhmTERF1在旱后复水花生根系恢复生长中的作用.结果表明,干旱抑制了花生根系的生长,根尖细胞死亡,AhmTERF1表达下调,mtDNA拷贝数降低.复水后,根系平均长度、平均表面积、平均直径和平均体积均得到恢复,根尖有新细胞长出,AhmTERF1表达和mtDNA拷贝数均增加.本研究可为后续认识干旱胁迫复水后花生根系的恢复生长调控机制奠定基础.Peanut is an important economic crop and oil crop in our country.In order to study the changes of physiological characteristics of peanut roots during drought and rehydration,peanut seedlings were subjected to drought treatment,and the growth of peanut roots was observed after rehydration,and the changes of root growth related indexes were analyzed.Trypan blue staining was used to detect the survival status of root tip cells.The expression changes of AhmTERF1 were further analyzed to explore the role of AhmTERF1 in peanut root growth under drought and after rehydration recovery.The results showed that drought stress inhibited the growth of peanut roots,resulted in cell death in root tips and decreasedAhmTERF1 expression and mtDNA copy number.After rehydration,the average length,average surface area,average diameter and average volume of peanut roots were recovered.New cells grow at the root tip and AhmTERF1 expression and mtDNA copy number were increased.This study lays a foundation for further understanding the regulation mechanisms of peanut roots under drought and after rehydration recovery.
