缩泉润肠方通过促进SCF、c-Kit表达改善小鼠老年性便秘

摘　　要：【目的】观察缩泉润肠方对老年性便秘小鼠的治疗作用及机制。【方法】将老年昆明小鼠随机分为便秘组、缩泉润肠方组、ISCK03[干细胞因子(SCF)、酪氨酸激酶生长因子受体(c-Kit)信号通路抑制剂]组、缩泉润肠方+ISCK03组、正常组,每组12只。除正常组外,其他组老年昆明小鼠均通过灌胃150 mg/kg复方地芬诺酯混悬液构建便秘模型。建模成功后,各组进行相应干预2周。干预结束后,测定小鼠粪便粒数、粪便含水量,酶联免疫吸附分析(ELISA)检测血清中白细胞介素(IL)-1β、IL-6、胃动素、胃泌素、5-羟色胺(5-HT)、血管活性肠肽(VIP)水平,测定小鼠胃排空率和小肠推进率,苏木精-伊红(HE)染色法观察结肠组织的病理变化,Western Blot法检测结肠组织中SCF、c-Kit蛋白表达。【结果】与正常组比较,便秘组小鼠结肠组织的细胞结构被破坏,肌肉层变薄,黏膜上皮出现损伤,粪便粒数,粪便含水量,血清中胃动素、胃泌素、5-HT水平,胃排空率、小肠推进率,结肠组织中SCF、c-Kit蛋白表达水平降低,血清中IL-1β、IL-6、VIP水平升高,差异均有统计学意义(P<0.05);与便秘组比较,缩泉润肠方组小鼠结肠组织损伤有所改善,粪便粒数、粪便含水量,血清中胃动素、胃泌素、5-HT水平,胃排空率、小肠推进率,结肠组织中SCF、c-Kit蛋白表达水平升高,血清中IL-1β、IL-6、VIP水平降低,差异均有统计学意义(P<0.05),而ISCK03组小鼠对应指标变化趋势与缩泉润肠方组相反(P<0.05);与缩泉润肠方组比较,缩泉润肠方+ISCK03组小鼠结肠组织损伤加剧,粪便粒数,粪便含水量,血清中胃动素、胃泌素、5-HT水平,胃排空率、小肠推进率,结肠组织中SCF、c-Kit蛋白表达水平降低,血清中IL-1β、IL-6、VIP水平升高,差异均有统计学意义(P<0.05)。【结论】缩泉润肠方可有效改善小鼠老年性便秘,其治疗作用与上调SCF、c-Kit表达,进而促进肠蠕动有关�Objective To observe the therapeutic effect of Suoquan Runchang Prescription for senile mice with constipation and its mechanism of action.Methods Senile Kunming mice were randomly divided into constipation group,Suoquan Runchang Prescription group,ISCK03[stem cell factor(SCF),receptor tyrosine kinase(c-Kit)signaling pathway inhibitor]group,Suoquan Runchang Prescription+ISCK03 group,and normal group,with 12 mice in each group.Except for the normal group,a constipation model was constructed by gavage of 150 mg/kg Compound Diphenoxylate Suspension in all groups of senile Kunming mice.After successful modeling,each group was treated for two weeks.At the end of the intervention,the fecal particles count and fecal water content were detected,the serum levels of interleukin(IL)-1β,IL-6,motilin,gastrin,5-hydroxytryptamine(5-HT),and vasoactive intestinal peptide(VIP)were detected by enzyme-linked immunosorbent assay(ELISA),and the changes in gastric emptying rate and small intestine propulsion rate were determined,the pathologic changes of colonic tissues were detected by hematoxylin-eosin(HE)staining,and protein expressions of SCF and c-Kit in the colonic tissues was detected by Western Blot.Results Compared with the normal group,the disrupted cellular structure,thinner muscle layer,damaged mucosal epithelium were seen in the constipation group,and the fecal particle count,fecal water content,serum motilin,gastrin and 5-HT levels,gastric emptying rate,small intestine propulsion rate,the protein expression levels of SCF and c-Kit in colonic tissues were decreased,and the serum IL-1β,IL-6 and VIP levels were increased,the differences being statistically significant(P<0.05).Compared with the constipation group,the colonic tissues damage were improved in Suoquan Runchang Prescription group,the fecal particle count,fecal water content,serum motilin,gastrin and 5-HT levels,gastric emptying rate,small intestine propulsion rate,protein expression levels of SCF and c-Kit in colonic tissues were elevated,the serum IL-1β,IL-6 a
