富硒植物乳植杆菌SAL干预衰老小鼠氧化应激损伤及肠道菌群失调

摘  要:目的探讨富硒植物乳植杆菌SAL对D−半乳糖所致衰老小鼠模型氧化应激损伤及肠道菌群失调的影响。方法将32只SPF级KM雄性小鼠均分为正常对照组(CON组)、模型组(MOD组)、亚硒酸钠干预组(SE组)和富硒植物乳植杆菌SAL干预组(SE-SAL组)。除CON组外,其余3组小鼠均用D−半乳糖构建衰老模型。SE组和SE-SAL组小鼠每日分别给予亚硒酸钠和富硒SAL株菌悬液灌胃,MOD组和CON组小鼠给予同等剂量的生理盐水灌胃,持续10周。收集各组小鼠新鲜粪便进行宏基因组测序分析,同时收集血清与肝脏组织进行硒水平、衰老指标β-半乳糖苷酶和氧化应激指标的检测,取结肠组织进行形态学检查,基于属水平的差异菌群与血清和肝脏氧化应激指标进行Spearman相关性分析。结果与CON组和MOD组比较,SE组和SE-SAL组小鼠血清与肝脏的硒水平均升高(t=15.485、19.426,均P<0.05;t=24.452、22.804,均P<0.05;t=19.864、21.495,均P<0.05;t=28.457、23.445,均P<0.05);与MOD组比较,SE组和SE-SAL组小鼠的β-半乳糖苷酶活性均下降(t=9.704、12.965,均P<0.05),SE-SAL组小鼠血清和肝组织MDA水平均降低(t=11.472、7.021,均P<0.05),SOD、GSH-Px、CAT和T-AOC活性均升高(t=12.520、23.047、8.317、8.425,均P<0.05;t=30.524、19.486、18.452、7.395,均P<0.05);与SE组比较,SE-SAL组小鼠的血清CAT和T-AOC活性均升高(t=6.157、6.846,均P<0.05);SE组和SE-SAL组小鼠结肠组织结构有所改善,其中,SE-SAL组小鼠结肠组织结构明显改善,隐窝数量增多且结构恢复,黏膜下层间隙变小且规则;SE-SAL组小鼠肠道Firmicutes和Verrucomicrobia丰度增加,Bacteroidetes丰度降低,F/B值升高(Z=3.964,P<0.05),显著差异的菌属Lactobacillus、Bifidobacterium和Turicibacter与MDA水平呈显著负相关(r=−0.682、−0.695、−0.805,均P<0.05),与SOD、GSH-Px、T-AOC活性呈显著正相关(r=0.738、0.751、0.790,均P<0.05;r=0.745、0.687、0.913,均P<0.05;r=0.816、0.686、0.789,均P<0.05)。结论富硒Objective To observe the effects of selenium-enriched Lactiplantibacillus plantarum SAL on oxidative stress injury and intestinal microbiota dysbiosis in aging mice.Methods A total of 32 male SPF KM mice were equally divided into four groups:normal control group(CON group),model group(MOD group),sodium selenite intervention group(SE group)and selenium-enriched Lactiplantibacillus plantarum SAL intervention group(SE-SAL group).Except for the CON group,the other three groups were established aging models with D-galactose.Then,the SE group and SE-SAL group were given sodium selenite and selenium-enriched Lactiplantibacillus plantarum SAL suspension respectively by gavage every day,while the MOD group and CON group were administered an equivalent dose of physiological saline.After ten weeks,the fresh feces of mice in each group were collected for metagenomic sequencing and analysis.Serum and liver tissues were collected to detect selenium content,senescence markers such asβ-galactosidase(β-Gal)and oxidative stress indicators,and colon tissue morphology was examined.Spearman correlation analysis was conducted to examine the correlation between differentially abundant microbiota at the genus level and oxidative stress indicators of serum and liver.Results Compared with the CON group and MOD group,the levels of Se in serum and liver in the SE group and SE-SAL group increased(all P<0.05).Compared with the MOD group,the activity ofβ-Gal in the SE group and SE-SAL group decreased(all P<0.05),the levels of MDA in serum and liver tissue in the SAL group decreased(all P<0.05),and the activities of SOD,GSH-Px,CAT and T-AOC increased(all P<0.05).The structure of colon tissue in the SE-SAL group showed marked improvement,with an increase in the number of crypts and restoration of their structure,and a smaller and more regular submucosal gap.The abundance of Firmicutes and Verrucomicrobia in the gut in the SE-SAL group increased,while that of Bacteroidetes decreased,and the F/B increased(P<0.05).The significantly different bac
