摘  要:目的探究龋齿儿童和健康儿童口腔菌群结构及多样性的差异,为该类患儿的治疗提供参考。方法应用16S rRNA基因V3−V4区高通量测序技术分析90名2~12岁儿童的口腔样本,对比其口腔菌群差异。结果儿童口腔存在核心微生物群,多个菌属的流行度大于90.00%,包括链球菌属(100.00%)、嗜血杆菌属(98.89%)、奈瑟菌属(97.78%)等。丰度前五的菌门均为厚壁菌门、变形菌门、放线菌门、拟杆菌门、梭杆菌门。龋齿儿童和健康儿童口腔菌群多样性指数差异有统计学意义(P<0.05)。LEfSe分析表明,龋齿儿童和健康儿童口腔无色杆菌属、假单胞菌属、约翰森菌属等的丰度差异有统计学意义(均P<0.05)。KEGG通路预测表明,龋齿儿童口腔菌群在免疫系统、形成与再生、细胞运动、聚糖的生物合成和代谢等通路上富集程度上升。结论龋齿儿童与健康儿童口腔菌群的结构、物种多样性以及群落功能均存在显著差异。Objective To explore the differences in community structure and diversity of oral flora between dental caries children and healthy children,providing a reference for the treatment.Methods High-throughput sequencing in the V3−V4 region of 16S rRNA gene was used to test the oral flora samples of 90 children aged 2−12 years.The difference of oral flora was compared.Results There was a core microbiota in children's oral cavity,and the prevalence of many genera were higher than 90.00%,including Streptococcus(100.00%),Haemophilus(98.89%),Neisseria(97.78%),etc.The top five phyla in terms of abundance were Firmicutes,Proteobacteria,Actinomycetes,Bacteroidetes and Fusobacterium.There was a significant difference in the oral microbiota diversity index between dental caries children and healthy children(P<0.05).LEfSe analysis showed that there were significant differences between the two groups in the abundances of Achromobacterium,Pseudomonas and Johnsonella(all P<0.05).The KEGG pathway prediction indicated that the dental caries children had an increased enrichment in metabolic pathways such as immune system,formation and regeneration,cell movement,polysaccharide biosynthesis and metabolism.Conclusion There are significant differences in the structure,species diversity,and functional changes of oral flora between the dental caries children and healthy children.
