舒节合剂对急性痛风性关节炎(湿热蕴结证)大鼠RKIP/NLRP3信号通路的影响

摘　　要：目的探究舒节合剂治疗急性痛风性关节炎(湿热蕴结证)大鼠的效果及机制。方法将雄性SD大鼠随机分为空白对照组、模型组、秋水仙碱片组和舒节合剂组,每组10只。空白对照组正常喂养,模型组、秋水仙碱组、舒节合剂组采用双重造模方法制备大鼠湿热蕴结型痛风性关节炎模型。造模成功后舒节合剂组给予舒节合剂浓缩液灌胃(18.81 mg/kg),秋水仙碱组给予秋水仙碱溶液(9 mg/kg)灌胃,空白对照组、模型组给予等体积生理盐水灌胃。观察造前后大鼠关节肿胀程度、关节炎症指数,检测血清中Raf激酶抑制剂蛋白(RKIP)、NOD样受体热蛋白结构域相关蛋白3(NLRP3)、凋亡相关斑点样蛋白(ASC)、半胱天冬蛋白酶-1(CASP-1)、白细胞介素-1β(IL-1β)的水平变化。结果经舒节合剂方剂干预后,模型大鼠的关节肿胀度减轻、炎症指数评分降低。舒节合剂组大鼠血清中RKIP水平升高(P<0.05),NLRP3、ASC、CASP-1及IL-1β水平显著降低(P<0.05)。结论舒节合剂可能通过RKIP/NLRP3信号通路抑制NLRP3炎性小体激活,减少IL-1β产生,进而发挥治疗痛风性关节炎的作用。Objective:To investigate the efficacy and mechanism of Shujie Mixture in treating rats with acute gouty arthritis(dampness-heat amassment pattern).Methods:Male SD rats were randomly divided into four groups:control group(blank control),model group,colchicine tablet group,and Shujie Mixture group,with 10 rats in each group.The control group was normally fed,while the model group,colchicine group,and Shujie Mixture group were subjected to a dual-modeling method to establish a rat model of gouty arthritis with dampness-heat amassment pattern.After successful modeling,the Shujie Mixture group was administered a concentrated solution of Shujie Mixture by gavage(18.81 mg/kg),the colchicine group was given colchicine solution by gavage(9 mg/kg),and the control and model groups were given an equal volume of normal saline by gavage.Before and after modeling,the degree of joint swelling and joint inflammation index in the rats were observed.Levels of Raf kinase inhibitor protein(RKIP),NOD-like receptor pyrin domain-containing protein 3(NLRP3),apoptosis-associated specklike protein(ASC),cysteinyl aspartate-specific protease(CASP-1),and interleukin(IL)-1βin serum were detected.Results:After intervention with Shujie Mixture,the degree of joint swelling and inflammatory index scores in the model rats were significantly reduced.In the Shujie Mixture group,the serum levels of RKIP increased(P<0.05),while the levels of NLRP3,ASC,CASP-1,and IL-1βsignificantly decreased(P<0.05).Conclusion:Shujie Mixture may inhibit the activation of the NLRP3 inflammasome through the RKIP/NLRP3 signaling pathway,thereby reducing the production of IL-1βand exerting therapeutic effects on gouty arthritis.
