黄精多糖对胃溃疡大鼠胃黏膜的保护作用及作用机制

摘  要:目的:探讨黄精多糖对胃溃疡(GU)大鼠胃泌素(GAS)、生长抑素(SS)、炎症因子及氧化水平的影响。方法:将50只胃溃疡大鼠随机分为模型组、黄精多糖(低、中、高)剂量组、奥美拉唑组,每组各10只。除空白组、模型组外,其余组给予相应药物灌胃8周后取大鼠胃组织。采用ELISA法检测大鼠GAS、SS、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、超氧化物歧化酶(SOD)活力、丙二醛(MAD)、谷胱甘肽过氧化物酶(GSH-Px)含量。结果:与空白组比,模型组黄精多糖低剂量组大鼠胃组织SS降低(F=271.066,P<0.05),胃泌素(GAS)、IL-6、TNF-α升高(F=278.373、164.100、115.230,P<0.05),SOD、GSH-Px降低(F=279.723、87.303,P<0.01)。结论:黄精多糖有抗炎抗氧化、保护胃溃疡大鼠胃黏膜的作用,其机制与上调SS、SOD、GSH-Px,下调GAS、MDA、IL-6、TNF-α的含量有关。Objective:To investigate the effects of Polygonatum polysaccharides on gastrin(GAS),somatostatin(SS),inflammatory factors,and oxidative levels in Gastric Ulcer(GU)rats.Methods:50 rats with gastric ulcers were randomly divided into a model group,a low,medium,and high dose group of Polygonatum polysaccharide,and an omeprazole group,with 10 rats in each group.Except for the blank group and the model group,the other groups were given corresponding drugs by gavage for 8 weeks.Gastrohormone(GAS),growth hormone inhibitor(SS),interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),superoxide dismutase(SOD)activity,malondialdehyde(MAD)and glutathione peroxidase(GSH-Px)contents were detected by ELISA.Results:Compared with the blank group,SS decreased(F=271.066,P<0.05),gastrin(GAS),IL-6 and TNF-αincreased(F=278.373,164.100,115.230;P<0.05),SOD and GSH-Px were significantly decreased(F=279.723,87.303;P<0.01)in model group.Conclusion:Huangjing polysaccharide has anti-inflammatory and antioxidant effects,as well as protective effects on gastric mucosa in ulcerated rats.Its mechanism is related to upregulating SS,SOD and GSH-Px,and downregulating GAS,MDA,IL-6 and TNF-α.
