摘  要:目的 探讨水溶性纤维素通过调控肠道菌群对大鼠IR的影响。方法 24只雄性SD大鼠随机分为正常对照(NC)组、高脂日粮(HFD)组、水溶性纤维(F-HFD)组,每组8只。检测各组体重、BG及血清支链氨基酸(BCAAs)、FIns,进行糖耐量试验,计算ISI、胰岛素抵抗指数(HOMA-IR),采用16Sr DNA基因扩增和测序确定肠道微生物组成。结果 NC、F-HFD、HFD组FPG、HOMA-IR、BCAAs依次升高(P<0.05),HFD组FIns高于NC、F-HFD组(P<0.05),ISI低于NC、F-HFD组(P<0.05)。与NC组比较,HFD组粪便中普雷沃菌(Prevotella)、拟杆菌门(Bacteroides)相对丰度升高(P<0.05),罗氏菌(Roseburia)降低(P<0.05)。与HFD组比较,F-HFD组Prevotella相对丰度降低(P<0.05)。相关性分析显示,BCAAs与IR、葡萄糖耐受、Prevotella呈正相关(P<0.05)。结论 水溶性纤维素可能通过下调高脂饮食大鼠肠道菌群Prevotella丰度,降低BCAAs生成,改善IR。Objective To investigate the correlation between water-soluble fibre and insulin resistance(IR)through regulating intestinal flora.Methods 24 male SD rats were randomly divided into normal control(NC)group,high-fat diet(HFD)group,and mixed 10%water-soluble fibre(F-HFD)group,with eight rats in each group.Body weight,blood glucose,serum branch chain amino acids(BCAAs)and FIns were measured.Glucose tolerance test and insulin tolerance test were performed.ISI and assessment-insulin resistance(HOMA-IR)was calculated.16SrDNA gene amplification and sequencing were performed to determine the composition of gut microorganisms in the intestinal contents of the rats.Results The FPG,HOMA-IR,and BCAAs were sequentially increased.FIns was higher in HFD group than in NC and F-HFD groups(P<0.05),ISI was lower in HFD group than in NC and F-HFD groups(P<0.05).The relative abundance of Prevotella and Bacteroides in feces was significantly higher(P<0.05)and Roseburia was significantly lower(P<0.05)in the HFD group compared with the NC group.The relative abundance of Prevotella was decreased in the F-HFD group compared with the HFD group(P<0.05).Correlation analysis showed that BCAAs were positively correlated with IR,glucose tolerance and Prevotella(P<O.05).ConclusionsWater-soluble fibre may reduce the production of BCAAs by down-regulating the abundance of intestinal flora Prevotella in rats on a high-fat diet,leading to IR reduction.
