摘 要:目的:通过体内外实验研究探讨金丝桃苷降低血浆总胆固醇的能力及其作用机制。方法:喂食高脂高胆固醇饲料建立高胆固醇血症小鼠模型,金丝桃苷干预组及阳性对照组给予不同剂量金丝桃苷灌胃和洛伐他汀灌胃8周。记录各组小鼠体质量变化,检测小鼠血浆胆固醇相关指标含量,观察肝组织病理改变、脂质累积情况及肝细胞摄取低密度脂蛋白胆固醇(LDL-C)的能力,检测金丝桃苷干预后肝细胞和肝组织中低密度脂蛋白受体(LDLR)丰度变化。结果:高胆固醇血症小鼠模型建立成功。与高胆固醇模型组相比,金丝桃苷干预组小鼠血总胆固醇(TC)水平下降(P<0.05);低剂量金丝桃苷干预组LDL-C水平下降(P<0.05);高胆固醇模型组小鼠肝组织出现大量空泡脂肪样变,肝细胞稀疏且排列紊乱,金丝桃苷干预组小鼠较高胆固醇模型组小鼠肝组织脂肪样变明显减轻,脂滴数量显著减少;体外实验发现,金丝桃苷能增加肝细胞中TC和LDL-C的含量(P<0.01),促进肝细胞对LDL-C的摄取,提高肝细胞及小鼠肝组织中LDLR丰度(P<0.01)。结论:金丝桃苷可能通过提高LDLR丰度促进小鼠肝组织摄取血LDL-C,进而降低小鼠血浆胆固醇水平。Objective:To intestigate the ability and mechanism of action of hyperoside in reducing plasma total cholesterol levels through in vitro and in vivo experiments.Methods:The mice model with hypercholesterolemia was established by feeding them high-fat and high-cholesterol chow,the hypericin intervention group and positive control group received different doses of hyperoside and lovastatin by gavage for 8 weeks.The changes in weight of the mice in each group were recorded.The content of serum cholesterol related indicators in mice were measured.The pathological changes and lipid accumulation of hepatic tissues and the uptake capacity of the hepatocytes for low-density lipoprotein cholesterol(LDL-C)was observed.The abundance of low density lipoprotein receptor(LDLR)proteins in the hepatocytes and hepatic tissues was detected after the intervention of hyperoside.Results:The mice model of hypercholesterolemia was successfully established.Compared to the hypercholesterolemia model group,the mice of hypericin intervention group showed a significant decrease in blood total cholesterol(TC)levels(P<0.05),and low-density hypericin intervention group showed a significant decrease in LDL-C(P<0.05).The hepatic tissues of mice in the hypercholesterolemia model group exhibited a large number of vacuolated steatosis,and the hepatocytes were sparse and disorganized.The hepatic tissues steatosis and the number of lipid droplets was significantly reduced in the hypericin intervention group compared to thehypercholesterolemia model group.The vitro experiments showed that hyperoside significantly increased TC levels and LDL-C content in hepatocytes(P<0.01),promoted LDL-C uptake in hepatocytes and increased abundance of LDLR in hepatocytes and hepatic tissues of mice(P<0.01).Conclusions:Hyperoside may promote the uptake of LDL-C in the hepatic tissues of mice by increasing the abundance of LDLR,thereby reducing the plasma total cholesterol levels of mice.
