黄芪-莪术含药血清对肿瘤血管内皮细胞抑制作用的体外实验研究

摘  要:目的通过细胞实验,基于对肿瘤血管内皮细胞增殖、迁移及血管形成的影响,探讨黄芪-莪术配伍抑制肿瘤血管生成的作用。方法首先制备中药黄芪、莪术及黄芪-莪术含药血清,然后以经肺癌Lewis细胞体外培养液上清诱导后的小鼠肺微血管内皮细胞,作为肿瘤源性肺微血管内皮细胞进行体外干预,通过CCK-8法检测细胞增殖率、细胞划痕实验检测细胞迁移能力、体外血管形成能力检测,探讨黄芪-莪术含药血清在体外抑制肿瘤血管生成的作用。结果细胞相对增殖率,黄芪血清组、莪术血清组以及黄芪-莪术血清组均低于无药血清组,差异有统计学意义(P<0.05),黄芪-莪术血清组尤为显著(P<0.05);细胞迁移能力,黄芪血清组、莪术血清组、黄芪-莪术血清组划痕愈合率均显著低于无药血清组(P<0.05),黄芪-莪术血清组最显著,且差异有显著意义(P<0.05);体外成血管能力,各中药含药血清组体外管腔数量均显著少于无药血清组(P<0.05),其中黄芪-莪术血清组最少,且差异有显著意义(P<0.05)。结论黄芪-莪术含药血清可以抑制肿瘤血管内皮细胞的增殖、迁移及成管能力,且两药合用时抑制作用更强。Objective To investigate the inhibitory effects of Radix Astragali Seu Hedysari-Rhizoma Curcumae on tumor angiogenesis in general and the impact of this drug on the proliferation,migration,and tube formation of tumor vascular endothelial cells in particular.Methods Traditional Chinese medicine containing Radix Astragali Seu Hedysari-Rhizoma Curcumae,along with a serum derived from these ingredients,was prepared and used to intervene in vitro in Lewis lung cancer cells and tumor-derived lung microvascular endothelial cells.The effects of serum containing Radix Astragali Seu Hedysari-Rhizoma Curcumae on tumor angiogenesis were assessed by studying cell proliferation using the CCK-8 assay,migration using the cell scratch assay,and by observing in vitro tube formation.Results The relative cell proliferation rate in the Radix Astragali Seu Hedysari,Curcuma,and Radix Astragali Seu Hedysari-Rhizoma Curcumae groups was significantly lower than that of the drug-free group(P<0.05),with the Radix Astragali Seu Hedysari-Rhizoma Curcumae group showing the most significant reduction(P<0.05).The scratch healing rate was also significantly reduced in these groups compared to the drug-free group(P<0.05),with the Radix Astragali Seu Hedysari-Rhizoma Curcumae group exhibiting the most pronounced effect(P<0.05).The in vitro angiogenic ability,as measured by the number of lumens,was significantly lower in the Radix Astragali Seu Hedysari-Rhizoma Curcumae group than in the drug-free group(P<0.05).Conclusion Radix Astragali Seu Hedysari-Rhizoma Curcumae can inhibit the proliferation,migration,and tube-formation of tumor vascular endothelial cells with a synergistic inhibitory effect.
